摘要
以三白草为药材,采用微波辅助水提法提取其多糖,利用响应面法优化提取条件。提取液经过纯化得到精多糖。通过对α-葡萄糖苷酶的抑制率来研究其降血糖活性。结果表明,三白草多糖的最佳提取条件为液料比25 mL/g,时间90 s,微波功率460 W,此时得到的多糖提取率约为2.57%,优于普通水提法(0.9%)。通过DEAE和G-75柱纯化得到精多糖SP-3。SP-3的IC 50值为0.002 mg/mL,小于阿卡波糖(0.003 mg/mL),因此,三白草具有较好的降血糖活性,且高于阿卡波糖。
The Saururus chinensis polysaccharides were extracted by microwave-assisted water extraction.The extraction conditions were optimized by response surface methodology.The extract was purified to obtain fine polysaccharide.The hypoglycemic activity was studied by the inhibition rate ofα-glucosidase.The results showed that the optimal extraction conditions of Saururus chinensis polysaccharide were the liquid-solid ratio of 25 mL/g,the time of 90 s,and the microwave power of 460 W.The extraction rate of polysaccharide obtained under this condition was about 2.57%,which was higher than that of ordinary water extraction(0.9%).After passing through the DEAE and G-75 column to obtain refined polysaccharides.The IC50 value of SP-3 was 0.002 mg/mL,which was lower than that of acarbose(0.003 mg/mL).Therefore,Saururus chinensis had good hypoglycemic activity,and was higher than acarbose.
