黄地清肠丸治疗溃疡性结肠炎大鼠模型的疗效观察

The Effect Observation of Huangdi Qingchang Wan in the Treatment of Ulcerative Colitis:An Experimental Study on Rat’s Model

摘要 目的:探究黄地清肠丸治疗溃疡性结肠炎(UC)大鼠模型的效果。方法:将120只雄性SD大鼠分为正常组,模型组,低剂量组,中剂量组,高剂量组和康复新组6组,每组20只。除正常组外,其余组均采用三硝基苯磺酸构建UC大鼠模型。造模成功后,正常组和模型组给予生理盐水灌肠,康复新组给予康复新液灌肠,低、中、高剂量组分别给予黄地清肠丸20g/kg·d、40g/kg·d和80g/kg·d药物灌肠治疗,周期14d。治疗期间观察各组大鼠疾病活动指数(DAI),治疗后行肠道病理学、血清TNF-α、IL-4和IL-17检测。结果:康复新组和低、中、高剂量组,DAI分值均随时间的延长持续降低(P <0.05),治疗第7天与第14天,低剂量组与康复新组DAI分值相当(P>0.05),随着黄地清肠丸药物浓度的增加DAI分值越低(P <0.05)。治疗后,与正常组相比,模型组血清中TNF-α和IL-17含量显著升高(P <0.05),IL-4的含量显著降低(P <0.05);与模型组比较,康复新组和各剂量黄地清肠丸组治疗后能有效降低TNF-α和IL-17水平(P <0.05),升高IL-4水平(P <0.05)。低、中剂量组与康复组治疗效果相当(P>0.05),但高剂量组疗效明显优于康复新组(P <0.05)。病理学分析显示,康复新和黄地清肠丸对肠道组织创面均有修复效果,各组间差异无统计学意义(P>0.05)。结论:黄地清肠丸能够有效降低UC大鼠模型血清中TNF-α和IL-17水平,升高IL-4水平,改善疾病症状,修复肠道病变组织。 Objective To investigate the effect of Huangdi Qingchang Wan(a TCM formula,Wan) in the treatment of ulcerative colitis(UC) in rat’s model.Methods Divided 120 male SD rats as normal group,model group,low-dose group,mid-dose group and high-dose group,and Kangfuxin group,20 rats for each;except for normal group,by using TNBS(trinitro-benzene-sulfonic acid) other groups were estabilised as UC rat’s model,the successfully established model groups received follwing enema therapy as:normal group and model group,normal saline for enema;Kangfuxin group,Kangfuxin liquid for enema;low-dose group,Wan of 20 g/kg·d for enema;mid-dose group,Wan of 40 g/kg·d for enema;high-dose group,Wan of 80 g/kg·d for enema,all groups’ treatment lasted 14 days;during this duration all groups’ DAI were observed,as well as after treatment all group received intestinal patological exam.,and the detection of serum TN F-α,IL-4 and IL-17.Results In Kangfuxin group as well as low,mid,and high-dose groups DAI point-value continously declined with time went(P <0.05);on the 7 th and 14 th day of treatment DAI pointvalue of low-dose group and Kangfuxin group were about equal(P>0.05), then, more Wan concentrations were,lower the point-value was(P <0.05).After treatment,compared to normal group model group’s serum TNF-α and IL-17 contents significantly uplifted(P <0.05),meanwhile,IL-4 one declined(P <0.05);compared to model group,Kangfuxin group and all three dose groups’ serum TNF-α and IL-17 contents effectively declined(P <0.05),meanwhile,IL-4 one uplifted(P <0.05).The treatment efficacy of low-and mid-dose group and Kangfuxin group were about equal(P>0.05),but the treatment efficacy in high-dose group was significantly better than that in Kangfuxin group(P <0.05).Pathological analysis showed Kangfuxin liquid and Wan had repair efficacy on intestinal tissue’s wound-surface,but there was no statistical difference among all groups(P>0.05).Conclusion Wan can effectively decrease rat’s serum levels of TNF-α and IL-17,increase IL-4 level,improve symptoms,and repair the lesion of intestinal tissue.
