人体肠道菌群对二甲基亚砜的体外降解作用

Degradation of DMSO by human gut microbiota through in vitro fermentation

摘要 目的通过二甲基亚砜(DMSO)与肠道菌群相互作用机制的研究,为DMSO的降解及其在药物研究中的应用提供肠道菌群方面的理论依据。方法采用分批发酵、气相色谱、气质联用和高通量测序技术研究DMSO的降解量与肠道菌群结构间的关系。结果发酵液中主要检测到甲硫醚、二甲基砜、二甲基二硫醚及甲基硫代磺酸甲酯等DMSO相关代谢产物;与其他他汀相比,生理浓度氟伐他汀组菌群的DMSO降解效果最佳,可能与Proteobacteria及Solobacterium和Escherichia-Shigella等菌群的相对丰度增加有关。不同肠道菌群产二甲基硫醚(DMS)水平差异明显,DMS分解可能与硫代谢途径相关,涉及的微生物主要是Desulfovibrio。结论人体肠道菌群可降解DMSO,其降解效率可能与Escherichia-Shigella的相对丰度有关,降解产物对人体肠道菌群结构无影响。 Objective To explore the mechanism of the interaction between dimethyl sulfoxide(DMSO)and human gut microbiota,and provide a theoretical basis for the degradation of DMSO and its application in drug research.Methods Batch fermentation,gas chromatography,gas chromatography-mass spectrometry and high-throughput sequencing were used to study the relationship between the degradation rate of DMSO and the changes of communities of gut microbiota.Results The main components in the fermentation broth were methyl sulfide,dimethyl sulfone,dimethyl disulfide,and S-methyl methanethiosulfonate.DMSO was best degradated in the broth containing physiological concentration of fluvastatin,which may be related to Proteobacteria,Solobacterium and Escherichia-Shigella.The amounts of DMS produced by different gut microbiota were significantly different.The degradation of DMS may be mainly related to the sulfur metabolism pathway and Desulfovibrio.Conclusion DMSO can be degraded by human gut microbiota;the degradation rate may be related to the relative abundance of Escherichia-Shigella,and the metabolites of DMSO has little effect on the structure of human gut bacterial community.
