骨形态发生蛋白15(BMP15)通过Smads信号通路促进雏鸡原始卵泡的激活 被引量:1

摘要 骨形态发生蛋白15(BMP15)在卵巢发育中发挥着重要的调节作用,然而,BMP15对雏鸡原始卵泡发育的作用尚不清楚。本试验研究了BMP15对雏鸡原始卵泡发育的影响。结果表明,BMP15和BMP受体-IB(BMPR1B)mRNA的表达变化以及BMPR1B蛋白的表达变化时间点与原始卵泡的激活时间点一致。BMP15通过促进卵母细胞和颗粒细胞的增殖和抑制细胞的凋亡来激活原始卵泡的活化,这种效应可被BMPR1B的抑制剂ML347所抑制(P<0.05)。电镜观察表明,BMP15处理可增加卵泡内卵母细胞与颗粒细胞之间的缝隙连接。与此同时,Smad1、Smad5和Smad4信号通路参与了BMP15的作用过程。以上结果表明,BMP15通过Smad1、Smad5和Smad4信号通路参与雏鸡原始卵泡的激活。 Bone morphogenetic proteins(BMPs),especially BMP15,plays essential roles in the regulation of ovarian development.However, the effect of BMP15 on the development of chicken primordial follicles is still unclear.This experiment was conducted to study the effects of BMP15 on primordium follicle development in chicks.The changes of expression of BMP15 and BMP receptor-Ib(BMPR1 B) mRNA and BMPR1 B protein were consistent with the activation time of primordial follicles.BMP15 activated primordial follicle activation by enhancing proliferation of oocytes and granulosa cells and reducing cell apoptosis.This effect was inhibited by a BMPR1 B inhibitor ML347(P<0.05).Electron microscopy showed that BMP15 increased the gap junctions between the oocytes and granulosa cells within the follicles.Meanwhile, Smad1,Smad5 and Smad4 signaling pathways were involved in the process of BMP15.These results indicated that BMP15 enhanced activation of primordial follicles in chickens through the Smad1,Smad5 and Smad4 signaling pathways.
