传染性法氏囊病毒VP2蛋白嵌合新城疫病毒样颗粒的构建和鉴定 被引量:1

Construction and identification of chimeric Newcastle disease virus-like particles with VP2 protein of infectious bursal virus

摘要 将传染性法氏囊病毒(infectious bursal disease virus, IBDV)强毒株的保护性蛋白VP2替换新城疫病毒(NDV)血凝素-神经氨酸酶(HN)蛋白胞外域部分,利用杆状病毒表达系统,构建了含有IBDV候选抗原表位VP2蛋白的重组杆状病毒rBV-VP2,并通过间接免疫荧光试验鉴定杆状病毒蛋白表达正确。rBV-VP2与NDV-HN、NDV-F、NDV-M组装成病毒样颗粒(cVLPs),形成含有IBDV候选抗原表位的IBD-ND二联病毒样颗粒(IBD-ND cVLPs),利用Western blot鉴定cVLPs各组分蛋白,证明IBD-ND cVLPs各组分组装正确。IBD-ND cVLPs的构建可为我国养禽业提供一种绿色、安全且可一针多防的IBD-ND二联病毒样颗粒疫苗候选株。 The ectodomain part of NDV hemagglutinin neuraminidase(HN) protein was replaced by the protective protein VP2 of the virulent strain of infectious bursal disease virus(IBDV).Using the baculovirus expression system, a recombinant baculovirus rBV-VP2 containing the candidate antigen epitope VP2 of infectious bursal disease virus(IBDV) was constructed, the correct expression of baculovirus protein was identified through indirect immunofluorescence test.rBV-VP2,NDV-HN,NDV-F,and NDV-M were assembled into cVLPs to form infectious bursal disease-Newcastle disease virus-like particles(IBD-ND cVLPs) containing infectious bursal disease candidate epitopes.The proteins of each component of cVLPs were identified by Western blot, which proved that the IBD-ND cVLPs were grouped correctly.The successful construction of the IBD-ND cVLPs will provide a green, safe and multi-dose vaccine candidate strain of infectious bursal disease and Newcastle disease combined virus-like particle vaccine for Chinese poultry industry.
