摘要
将辣木籽分别配制成5mg/mL、25mg/mL、125mg/mL、250mg/mL溶液,依次用石油醚、乙酸乙酯、正丁醇及水对其进行萃取。以阿卡波糖为阳性对照,用对硝基苯酚吡喃葡萄糖苷(PNPG)法测定辣木籽提取物降血糖能力。将辣木籽石油醚、乙酸乙酯、正丁醇及水提取物分别配成2mg/mL、4mg/mL、6mg/mL、8mg/mL、10mg/mL溶液,以考来烯胺为阳性对照,测定其胆酸盐的结合能力。辣木籽降血糖测试结果显示,辣木籽水相、正丁醇相、乙酸乙酯相在250mg/mL时,对α-葡萄糖苷酶抑制率分别为99.5%、98.9%、93.3%,均高于阳性对照92.5%,石油醚提取物对α-葡萄糖苷酶无明显抑制作用。辣木籽胆酸盐结合实验结果表明,辣木籽石油醚相及水相提取物对水合胆酸钠和甘胆酸钠具有较强的结合能力。辣木籽正丁醇相提取物和水相提取物对牛胆酸钠结合能力较强。
Moringa oleifera seeds were extracted with petroleum ether, ethyl acetate, n-butanol and water successively,and prepared into 5mg/mL, 25mg/mL, 125mg/mL and 250mg/mL solutions, respectively. The hypoglycemic activity of Moringaspa seed extract was determined by p-nitrophenol glucopyranoside(PNPG) method, and the acarbose of 5 mg/mL was used as positive control. Moringa oleifera seeds were extracted with petroleum ether, ethyl acetate, n-butanol and water successively, and prepared into 2mg/mL、4mg/mL、6mg/mL、8mg/mL、10mg/mL, and the cholestyramine of 10mg/mL was used as positive control. The hypoglycemic test results in vitro showed that the α-glucosidase inhibition rates of water phase, n-butanol phase and ethyl acetate phase at 250mg/mL were 99.5%, 98.9% and 93.3%, respectively, which were higher than those of positive control(92.5%). Petroleum ether extract had no significant inhibition effect on α-glucosidase. The results showed that petroleum ether and aqueous extract of Moringa oleifera seeds had strong binding ability to sodium cholate hydrate and sodium glycholate. The n-butanol and aqueous extracts of moringa oleifera seeds had stronger binding ability to sodium taurate.
