摘要
目的优化西红花花瓣中山柰酚-3-O-槐糖苷的大孔吸附树脂纯化工艺。方法以吸附量、解吸量、解吸率为评价指标,采用静态吸附-解吸试验筛选大孔吸附树脂型号;采用单因素试验分别考察上样液pH值、上样浓度、上样体积、洗脱剂(乙醇)体积分数以及洗脱体积对目标成分吸附或解吸的影响。结果最优工艺条件为HPD-100型大孔吸附树脂,上样液pH=4,生药浓度为2.0 g/mL,上样体积为1 BV,洗脱剂为45%乙醇,洗脱体积为3 BV。经HPD-100型大孔吸附树脂富集后,山柰酚-3-O-槐糖苷纯度达到57.40%,是粗提物的8.61倍。结论该工艺条件稳定可行,可用于西红花花瓣中山柰酚-3-O-槐糖苷的富集分离。
Objective To optimize the purification condition of kaempferol-3-O-sophoroside from Crocus sativus petals by macroporous adsorption resin.Methods Taking adsorption capacity,desorption capacity and desorption ratio as evaluation indexes,the macroporous resin was selected by static adsorption-elution test.Single factor tests were adopted to investigate the effect of loading sample pH value,sample concentration,sample volume,volume fraction of eluting solvent(aqueous ethanol)and elution volume on adsorption or desorption of target component.Results The optimal technology conditions were as followed:HPD-100 macroporous adsorption resin was chosen,pH value of loading sample was 4,crude drug concentration was 2.0 g/mL,sample volume was 1 BV,eluting solvent was 45%aqueous ethanol,eluant volume was 3 BV.The purity of kaempferol-3-O-sophoroside after enrichment by HPD-100 macroporous adsorption resin reached 57.40%,which was 8.61 times of crude extract.Conclusion HPD-100 macroporous adsorption resin can effectively purify kaempferol-3-O-sophoroside from Crocus sativus petals under the optimized conditions which are stable and feasible.
