基于HPLC指纹图谱对不同基原茵陈的区分及各品种茵陈质量评价 被引量:3

Identification of Difference Origins of Artemisia Scoparia Herba Based on Fingerprint and the Quality Evaluation of Different Varieties of Samples

摘要 目的建立2种基原茵陈(滨蒿、茵陈蒿)HPLC指纹图谱和多成分含量测定方法,基于该方法对滨蒿和茵陈蒿进行区分,并对不同品种茵陈进行质量评价。方法采用HPLC法构建2种基原茵陈对照药材、16批茵陈药材指纹图谱并测定11种化学成分含量,采用“中药色谱指纹图谱相似度评价系统(2012版)”进行相似度分析、以SPSS 21.0软件进行聚类分析、以SIMCA14.1进行主成分(PCA)分析,全面对不同基原茵陈药材进行质量评价。结果建立了2种基原茵陈的指纹图谱,共确定23个共有峰,指认并测定了其中11个化学成分,筛选出了2种基原茵陈药材的特异峰(峰11、峰12),茵陈蒿各成分均明显高于滨蒿。16批茵陈指纹图谱的相似度为0.872~0.989,6批样品被鉴定为滨蒿,10批样品被鉴定为茵陈蒿,相似度评价、聚类分析和主成分分析结论一致。正交偏最小二乘法-判别分析(OPLS-DA)共筛选出了5个质量标志性成分。结论该研究建立的方法稳定可靠,可用于茵陈药材的基原判定及质量控制。 Objective To establish the HPLC fingerprints of 2 origins of Artemisia Scoparia Herba(Artemisia scoparia Waldst.et Kit.and Artemisia capillaris Thunb.)and determine multi-component content.The differences of Artemisia scoparia Waldst.et Kit.and Artemisia capillaris Thunb.are compared based on the fingerprint method and the quality of different varieties of Artemisia Scoparia Herba samples are evaluated.Methods Fingerprints of 2 origins and 16 batches of Artemisia Scoparia Herba and 11 chemical contents were determined by HPLC method.The similarity evaluation was carried out by“the Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System(2012 version)”,the cluster analysis was performed by SPSS 21.0,and the PCA analysis was performed by SIMCA14.1,all these methods were used to evaluate the quality of different origins of Artemisia Scoparia Herba comprehensively.Results The fingerprints of 2 origins of Artemisia Scoparia Herba were established,23 common peaks and 11 chemical components were identified and determined.The specific peaks of 2 origins of Artemisia Scoparia Herba were selected as 11 and 12.The components of Artemisia capillaris Thunb.were significantly higher than Artemisia scoparia Waldst.et Kit..The similarity of 16 batches of samples was 0.872~0.989.Six batches of samples were determined as Artemisia scoparia Waldst.et Kit.,10 batches of samples were determined as Artemisia capillaris Thunb.,the results of the principal component analysis and the cluster analysis were consistent with the similarity evaluation.The orthogonal partial least squares discrimination analysis screened 5 varied components.Conclusion The method established is stable and reliable,and provides reliable analysis methods for the quality control and origins identification of Artemisia Scoparia Herba.
