雌激素受体β基因多态性与非梗阻性无精子症关系的Meta分析

Association between estrogen receptorβgene polymorphism and non-obstructive azoospermia:a Meta-analysis

摘要 目的利用Meta分析系统评价雌激素受体β(ERβ)的Rsa I(rs1256049)和Alu I(rs4986938)位点基因多态性与非梗阻性无精子症(NOA)的相关性。方法检索国内外各数据库关于ERβ与NOA的相关文献,其中国外数据库包括PubMed、Medline、Embase、Web of Science、Cochrane Library,国内数据库包括中国知网、万方数据库,检索时间从建库至2020年10月1日。利用STATA 12.0软件计算优势比及95%置信区间(95%CI)评价其相关性,并进行异质性、敏感性和发表偏倚分析。结果共纳入5篇病例对照研究,其中病例组702例,对照组1323例。对于Rsa I(rs1256049)基因位点,G vs.A(OR=0.580,95%CI:0.380~0.900)、GG vs.AG+AA(OR=0.560,95%CI:0.360~0.860)、GG vs.AG(OR=0.569,95%CI:0.374~0.894)三种模型结果均与NOA具有明显的相关性(均P<0.05),提示G等位基因是NOA的保护性因素,A等位基因可增加NOA风险,GG和GA基因型可降低NOA的风险。Alu I(rs4986938)基因多态性与NOA均无相关性(均P>0.05)。结论Rsa I(rs1256049)基因位点多态性与NOA具有明显的相关性,其中A等位基因可增加NOA风险,而Alu I(rs4986938)基因位点多态性与NOA无相关性。 Objective To systematically evaluate the association between Rsa I(rs1256049)and Alu I(rs4986938)polymorphisms of estrogen receptorβ(ERβ)and non-obstructive azoospermia(NOA)by Meta-analysis.Methods Literature on ERβand non-obstructive azoospermia was retrieved from domestic and foreign databases,including PubMed,Medline,Embase,Web of Science,Cochrane Library,and China National Knowledge Infrastructure(CNKI)and Wanfang Database.The retrieval time was from the establishment of the database to October 1,2020.The odds ratio and 95%confidence interval were calculated by STATA 12.O software to evaluate the correlation.Heterogeneity,sensitivity and publication bias were analyzed.Results A total of 5 case-control studies were included,including 702 cases in the case group and 1323 cases in the control group.For Rsa I(rs1256049)gene locus,G vs.A(OR=0.580,95%CI:0.380-0.900),CG vs.AG+AA(OR=0.560,95%CI:0.360-0.860),GG vs.AG(OR=0.569,95%CI:0.374-0.894)were significantly correlated with NOA(allP<0.05),suggesting that G allele was a protective factor for NOA,A allele could increase the risk of NOA,and GG and GA genotypes could reduce the risk of NOA.There was no correlation between Alu I(rs4986938)gene polymorphism and NOA(all P>0.05).ConclusionsThe polymorphism of Rsa I(rs1256049)gene locus is significantly correlated with NOA,among which the A allele can increase the risk of NOA,while the Alu I(rs4986938)gene polymorphism has no correlation with NOA.
