摘要 本文旨在概述miR-122对肝内外组织脂质代谢的调控作用及具体分子机制。MiR-122为肝脏特异性高表达miRNA,与肝内外组织脂质代谢调控及体内血脂水平密切相关。拮抗miR-122可抑制肝内甾醇反应元件结合蛋白-1(sterol regulatory element binding protein-1,SREBP-1)介导脂质合成并同时促进脂质β氧化和脂肪分解,防止肝脏脂质过度累积及非酒精性脂肪肝病进展。肝源性miR-122亦可经血液循环影响外周脂质代谢。拮抗miR-122可抑制脂肪细胞成脂肥大、改善血脂水平保护心肌细胞及促进骨骼肌纤维甘油三酯合成与储存,抑制体内肥胖症、动脉粥样硬化性心血管疾病及糖尿病的发展。总之,抑制miR-122表达及生物学作用可改善肝内外组织脂质代谢过程,防止脂代谢紊乱疾病的发生发展。 This paper aims to summarize the regulatory effect and specific molecular mechanism of miR-122on lipid metabolism in intra-hepatic and extra-hepatic tissues. MiR-122 is a liver-specific and highly expressed microRNA, which is closely related to the regulation of lipid metabolism inside and outside the liver and the level of blood lipids in vivo. Antagonizing miR-122 inhibits intrahepatic sterol regulatory element binding protein-1(SREBP-1)-mediated lipid synthesis and simultaneously promotes lipid β-oxidation and lipolysis,preventing excessive lipid accumulation in the liver and progression of nonalcoholic fatty liver disease. Liverderived miR-122 can also affect peripheral lipid metabolism through blood circulation. Antagonizing miR-122can inhibit adipogenesis and hypertrophy of adipocytes, improve blood lipid levels, protect cardiomyocytes,and promote the synthesis and storage of triglyceride in skeletal muscle fibers, thereby inhibiting the development of obesity, atherosclerotic cardiovascular disease and diabetes in vivo. In conclusion, inhibiting the expression and biological function of miR-122 can improve the lipid metabolism inside and outside the liver and prevent the occurrence and development of lipid metabolism disorders.
