摘要
目的探讨丹参酮IIA对脓毒症大鼠肺损伤的保护作用。方法采用盲肠结扎穿孔法(CLP),随机将50只健康成年雄性SD大鼠分为假手术组、模型组、低剂量治疗组(丹参酮IIA 5 mg/kg)、中剂量治疗组(丹参酮IIA 10 mg/kg)、高剂量组(丹参酮IIA 20 mg/kg),每组各10只。假手术组:麻醉,开腹翻动肠道,关腹,2 h后腹腔注射0.9%氯化钠溶液(20 mL/kg),24 h后麻醉并腹主动脉采血,直至大鼠缺血而死。模型组及3个不同剂量的治疗组造模成功后2 h腹腔注射0.9%氯化钠溶液(20 mL/kg)或丹参酮IIA(5、10、20 mg/kg),24 h后,大鼠麻醉成功后,采集腹主动脉动脉血,行血气分析,计算PaO_(2)/FiO_(2)及ELISA法检测各组大鼠左肺下叶肺组织匀浆中XO、GSH-Px、MDA、SOD含量。结果与模型组相比,三种剂量的丹参酮ⅡA均可以改善动脉血气中PaO_(2)/FiO_(2)比值,促进SOD和GSH-Px水平升高,同时降低了XO、MDA的水平。但是只有高剂量组的丹参酮ⅡA对MDA、SOD以及GSH-Px产生了明显的影响(P<0.05),低剂量与中剂量组中的MDA、SOD以及GSH-Px无显著变化(P>0.05)。结论丹参酮ⅡA在脓毒症肺损伤大鼠中可起抗炎、清除氧自由基的保护作用。
Objective To investigate the protective effect of Tanshinone IIA on lung injury in septic rats.Methods Cecal ligation and perforation(CLP)was performed,50 healthy adult male SD rats were randomly divided into sham operation,model group,low-dose treatment group(Tanshinone IIA 5 mg/kg),medium-dose treatment group(Tanshinone IIA 10 mg/kg)and high-dose group(Tanshinone IIA 20 mg/kg),with 10 rats in each group.The SD rats in the sham operation group successively went through anesthesia,open abdomen,turn over the intestines,close the abdomen,intraperitoneal injection of normal saline(20 mL/kg)2 hours later,anesthesia and blood collection from abdominal aorta 24 hours later,until the rats died of ischemia.The model group and the other three treatment groups with different doses were intraperitoneally injected with normal saline(20 mL/kg)or Tanshinone IIA(5,10,20 mg/kg)2 hours after successful modeling.After 24 hours the rats were anesthetized successfully,the abdominal aortic artery blood was collected for blood gas analysis.PaO_(2)/FiO_(2) was calculated.The contents of XO,GSH Px,MDA and SOD in the lung tissue homogenate of the lower lobe of the left lung were detected by ELISA.Results Compared with the model group,PaO_(2)/FiO_(2) in arterial blood gas increased,XO and MDA decreased,SOD and GSH PX increased in low-dose treatment group(Tanshinone IIA 5 mg/kg),medium-dose treatment group(Tanshinone IIA 10 mg/kg)and high-dose group(Tanshinone IIA 20 mg/kg)(P<0.05).However,there was no significant difference in the contents of MDA,SOD and GSH PX between the low-dose Tanshinone IIA treatment group and the medium dose Tanshinone IIA treatment group(P>0.05).Conclusion Tanshinone IIA has the protective effects of anti-inflammatory and scavenging oxygen free radicals in rats with sepsis lung injury.
