摘要
目的:通过网络药理学的方法筛选香加皮的主要活性成分,构建药物-活性成分-靶点-疾病网络,探讨香加皮治疗心力衰竭的机制。方法:运用中药系统药理学数据库与分析平台(TCMSP)、Uniprot及GeneCard等数据库获取香加皮的主要活性成分及其相关靶点、心力衰竭的靶点。通过String平台和Cytoscape 3.7.2软件绘制香加皮-活性成分-靶点-心力衰竭网络并构建蛋白质-蛋白质相互作用网络。利用Metascape和微生信平台进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:筛选到香加皮的19个有效成分及101个靶点,其中白蛋白(Albumin,ALB)、β肌动蛋白(β-Actin,ACTB)、过氧化物酶体增殖物激活受体γ(Peroxisome Proliferative Activated Receptorγ,PPARG)、过氧化氢酶(Catalase,CAT)、半胱氨酸天冬氨酸蛋白酶3(Caspase 3,CASP3)、可能是关键靶点,靶点基因功能主要富集在血液循环、细胞凋亡等过程,其作用生物路径主要有糖尿病并发症中的晚期糖基化终末产物(Advanced Glycation End Products,AGE)-晚期糖基化终末产物受体(Advanced Glycation End Product Receptor,RAGE)信号通路、低氧诱导因子-1(Hypoxia Inducible Factor-1,HIF-1)信号通路、环磷酸腺苷(Cyclic Adenosine Monophosphate,cAMP)信号通路、丝裂原活化蛋白激酶(Mitogen-activated Protein Kinase,MAPK)信号通路等。结论:运用网络药理学方法探讨香加皮治疗心力衰竭的作用机制可为香加皮治疗心力衰竭提供些许理论依据。
Objective:To screen the main active components of Xiangjiapi(Periplocae Cortex)through network pharmacology,construct drug-actire component-target-disease network,and explore the mechanism of Xiangjiapi in the treatment of heart failure.Methods:TCMSP,Uniprot and Genecard databases were used to obtain the main active components and their related targets,as well as the targets of heart failure.Xiangjiapi actire component-target-heart failure network and protein-protein interaction network were constructed by String platform and Cytoscape 3.7.2 software.The Go enrichment analysis and KEGG pathway enrichment analysis were performed by Metascape and Micro bioinformatics platform.Results:A total of 19 active components and 101 targets were obtained,among which ALB,ACTB,PPARG,CAT,CASP3 may be the key targets.The function of target genes was mainly enriched in blood circulation and cell apoptosis,and the biological pathways mainly included AGE-RAGE signaling pathway in diabetic complications,HIF-1 signaling pathway,cAMP signaling pathway,and MAPK signaling pathway,etc..Conclusion:Using network methods to explore the mechanism of Xiangjiapi in the treatment of heart failure,provide theoretical basis for the treatment of heart failure.
