子宫内膜癌组织中紧密连接蛋白6、紧密连接蛋白9的表达及临床意义

Expression and clinical significance of CLDN6 and CLDN9 in endometrial cancer

目的分析子宫内膜癌(EC)患者癌组织中紧密连接蛋白6(CLDN6)、紧密连接蛋白9(CLDN9)的表达水平及临床意义。方法选取2016年6月至2018年7月于青岛大学附属青岛市中心医院行手术治疗的68例EC患者作为研究对象,术中取其癌组织及癌旁组织,另将同期57例因宫颈病变切除全子宫、全子宫加双附件的患者的正常子宫内膜组织作为对照。检测不同组织中CLDN6、CLDN9蛋白表达情况,分析CLDN6、CLDN9表达与患者临床病理特征及预后的关系,并采用Ualcan数据库分析EC组织中CLDN6、CLDN9 mRNA表达情况及二者水平对患者预后的影响。结果CLDN6、CLDN9高表达患者的生存率均低于CLDN6、CLDN9低表达患者(P<0.05);癌组织中CLDN6、CLDN9蛋白阳性率均高于癌旁组织及正常子宫内膜组织(P<0.05),CLDN6、CLDN9蛋白水平与国际妇产科联盟(FIGO)分期、病理分级及淋巴结转移有关(P<0.05),CLDN6、CLDN9阳性患者3年生存率均低于CLDN6、CLDN9阴性患者(P<0.05)。结论CLDN6、CLDN9在EC患者癌组织中呈高表达,二者水平可预测患者预后情况。

Objective To analyze the expression levels and clinical significance of claudin 6(CLDN6)and claudin 9(CLDN9)in cancer tissues of patients with endometrial cancer(EC).Methods 68 patients with EC who underwent surgical treatment in Qingdao Central Hospital,the Affiliated Hospital of Qingdao University from June 2016 to July 2018 were selected as the study objects.and the endometrial cancer tissues and adjacent tissues were collected during the operation.The normal endometrial tissues of 57 patients who underwent total hysterectomy or total hysterectomy with double adnexa due to cervical lesions during the same period were taken as control group.The protein expressions of CLDN6 and CLDN9 in different tissues were detected to analyze the relationship between the expressions of CLDN6 and CLDN9 and the clinicpathological characteristics and prognosis of patients,the Ualcan database was used to analyze the expression of CLDN6 and CLDN9 mRNA in endometrial cancer tissues and the influence of the two levels on the prognosis of patients.Results The survival rate of patients with high expression of CLDN6 and CLDN9 was lower than that of patients with low expression of CLDN6 and CLDN9,respectively(P<0.05);the positive rates of CLDN6 and CLDN9 proteins in endometrial cancer tissues were higher than those in adjacent tissues and normal endometrial tissues(P<0.05),the levels of CLDN6 and CLDN9 proteins were related to the international federation of gynecology and obstetrics(FIGO)staging,pathological grading and lymph node metastasis(P<0.05),the 3-year survival rate of CLDN6 and CLDN9-positive patients was lower than that of CLDN6 and CLDN9-negative patients(P<0.05).Conclusions CLDN6 and CLDN9 are highly expressed in endometrial cancer tissues,and the levels of both can detect the prognosis of patients.
