摘要
目的探讨脂多糖(LPS)对大鼠海马齿状回(DG)的神经发生的影响及机制。方法大鼠腹腔注射LPS(1 mg/ml)后,用免疫组织化学方法检测海马DG的颗粒下层5溴脱氧尿嘧啶(BrdU)的阳性细胞和诱发的钙接头蛋白分子Iba1蛋白的表达;用免疫荧光双标的方法检测BrdU和胶质纤维酸性蛋白(GFAP)、双皮质醇(DCX)的共表达;新生大鼠海马分离培养神经干细胞,采用免疫细胞法检测神经上皮干细胞蛋白(Nestin)的表达,检测LPS对神经前体细胞分化的影响和LPS受体Toll样受体(TLR)4的表达。结果LPS能使青少年雄性大鼠发育迟缓,体重增长延迟;急性抑制成年大鼠DG区的神经发生。结论LPS可能通过激活TLR4途径抑制神经前体细胞的增殖。
Objective To investigate the effect of lipopolysaccharide(LPS)on hippocampal dentate gyrus(DG)in rats and its mechanism.Methods After intraperitoneal injection of LPS(1 mg/ml),the expression of 5 bromodeoxyuracil(BrdU)-positive cells and induced calcutin molecules Iba1 protein in the subgranular layer of hippocampal DG were detected by immunohistochemistry.The co-expression of BrdU,glial fibrillary acidic protein(GFAP)and double-cortin(DCX)were detected by immunofluorescence dual labeling method.Neural stem cells were isolated and cultured from the hippocampus of newborn rats.The expression of neuroepithelial stem cell protein(Nestin)was detected by immune cell assay.The effect of LPS on the differentiation of neural progenitor cells and the expression of Toll-like receptor(TLR)4 were detected.Results LPS caused growth retardation and weight gain delay in adolescent male rats.Acute inhibition of neurogenesis in adult rats hippocampal DG.Conclusions LPS may inhibit the proliferation of neural progenitor cells by activating TLR4 pathway.
