链脲佐菌素腹腔注射制作大鼠2型糖尿病模型的方式与剂量研究

The Mode and Dosage of Streptozotocin Intraperitoneal Injection in Establishment of Type 2 Diabetes Rat Model

目的:探讨链脲佐菌素腹腔注射建立大鼠2型糖尿病模型的最佳方式和剂量。方法:选择成年雄性SPF级SD大鼠,高糖高脂饲料喂养4周后,采用不同方式和剂量(一次性注射50mg/kg;首次注射30mg/kg,后补注射35mg/kg)腹腔注射链脲佐菌素以建立大鼠2型糖尿病模型,观察不同方式和剂量链脲佐菌素腹腔注射建立2型糖尿病模型的成功率及大鼠死亡率。结果:高糖高脂饲料喂养4周后行链脲佐菌素50mg/kg一次性腹腔注射,成模率高达93.75%,血糖值达24.70±5.67mmol/L,但死亡率达65.00%,迫使实验终止。而首次注射30mg/kg链脲佐菌素,1周后再次注射35mg/kg的方法,成模率可达92.11%,8周后死亡率仅6.58%。结论:高糖高脂饲料喂养4周后分次腹腔注射30~35mg/kg链脲佐菌素,是获得稳定的大鼠2型糖尿病模型且具备高成模率、低死亡率的方法。

Objective:To study the optimal mode and dosage of streptozotocin intraperitoneal injection in establishment of type 2 diabetes rat model.Methods:SPF-rated adult male SD rats were chosen to establish type 2 diabetes model by different modes and doses(in a single injection of 50mg/kg;30mg/kg for the first injection and 35mg/kg for the second)of streptozotocin intraperitoneal injection after 4 weeks of high-sucrose and highlipids feeding,and observed the success rate of model building and rat mortality.Results:After 4 weeks of high-sucrose and high-lipids feeding,50mg/kg of streptozotocin was intraperitoneal injected in one shot,which turned out a result that the success rate was 93.75%and blood glucose value was 24.70±5.67mmol/L,but the rat mortality was 65.00%,which made the experiment has to be aborted.However,the mode of 30mg/kg streptozotocin for the first injection and 35mg/kg for the second after 1 week showed that the success rate was 92.11%,and the rat mortality after 8 weeks was only 6.58%.Conclusion:Fractionated intraperitoneal injection with 30~35mg/kg streptozotocin after 4 weeks of high-sucrose and high-lipids feeding is a method to obtain stable type 2 diabetes rat model with high success rate and low mortality.
