复发性脑胶质瘤组织TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达与细胞增殖侵袭恶性行为的关系及预测预后的效能 被引量:2

摘要 目的探讨复发性脑胶质瘤组织四跨膜超家族成员1蛋白(TM4SF1)mRNA、锌转运体1(ZnT1)mRNA、miR-542-3p表达与细胞增殖侵袭恶性行为的关系及预测预后的效能。方法选取2016年3月至2020年3月我院收治的169例复发性脑胶质瘤患者,根据6个月预后情况分为死亡组、生存组。比较2组基线资料、组织TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达,并比较不同TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达者脑胶质瘤WHO分级,采用Cox比例风险回归模型分析预后相关因素,受试者工作特征曲线(ROC)及ROC下面积(AUC)分析各指标预测预后的效能,卡普兰-迈耶(Kaplan-Meier,KM)生存曲线分析不同TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达水平者生存率。结果死亡组距离首次手术时间、脑胶质瘤WHO分级、肿瘤切除程度、TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达与生存组比较,差异有统计学意义(P<0.05);以2组TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达均值为分界进行分层,TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p高表达与低表达患者脑胶质瘤WHO分级比较,差异有统计学意义(P<0.05);Cox比例风险回归模型将距离首次手术时间、脑胶质瘤WHO分级、肿瘤切除程度控制后,TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p仍与患者预后相关(P<0.05);TM4SF1 mRNA+ZnT1 mRNA+miR-542-3p预测预后的AUC为0.938,大于单一指标预测(P<0.05);TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p高危患者患者生存率低于低危达者(P<0.05)。结论脑胶质瘤组织TM4SF1 mRNA、ZnT1 mRNA、miR-542-3p表达与细胞增殖侵袭恶性行为关系密切,是预后的主要影响因素,三者联合可为临床预测预后提供参考依据。 Objective To investigate the correlation between the expressions of transmembrane 4 super family 1(TM4SF1)mRNA,zinc transporter 1(ZnT1)mRNA,and miR-542-3p in recurrent glioma tissues and the malignant behavior of cell proliferation and invasion,and to explore its prognostic performance.Methods A total of 169 patients with recurrent glioma were divided into death group and survival group according to the 6-month prognosis.The baseline data and the expressions of TM4SF1 mRNA,ZnT1 mRNA and miR-542-3p were observed and compared between the two groups.The Cox proportional hazards regression model was used to analyze the prognostic factors,and the receiver operating characteristic curve(ROC)and the area under the ROC(AUC)were used to analyze the prognostic efficacy of various indicators.Kaplan-Meier(KM)survival curve was used to analyze the survival rate of patients with different expression levels of TM4SF1 mRNA,ZnT1 mRNA,and miR-542-3p.Results There were significant differences in the time from the first operation,the WHO classification of glioma,the degree of tumor resection,the expression levels of TM4SF1 mRNA,ZnT1 mRNA,and miR-542-3p between the two groups(P<0.05).The two groups were stratified by the mean expression of TM4SF1 mRNA,ZnT1 mRNA,and miR-542-3p,there was a significant difference in the WHO classification of glioma between patients with high expression of TM4SF1 mRNA,ZnT1 mRNA,and miR-542-3p and those with low expression(P<0.05).After the Cox proportional hazard regression model controlled the time to first operation,WHO classification of glioma,and degree of tumor resection,TM4SF1 mRNA,ZnT1 mRNA,and miR-542-3p were still related to the prognosis of patients(P<0.05).The prognostic AUC of TM4SF1 mRNA+ZnT1 mRNA+miR-542-3p was 0.938,which was greater than that by single index prediction(P<0.05).Moreover the survival rate of patients with high expression of TM4SF1 mRNA and ZnT1 mRNA was significantly lower than that of patients with low expression(P<0.05),however,the survival rate of patients with low expression of miR-542-3p was significantly lower than that of patients with high expression(P<0.05).Conclusion The expression levels of TM4SF1 mRNA,ZnT1 mRNA,and miR-542-3p in glioma tissue are closely related to cell proliferation,invasion and malignant behavior,which is the main influencing factors on the prognosis of patients,and the combination detection of the three indexes can provide a reference for clinical prediction of prognosis.
