慈莲胶囊治疗实验性乳腺增生大鼠作用研究

Research on the Effect of Cilian Capsule in the Treatment of Experimental Mammary-gland Hyperplasia in Rats

目的探讨慈莲胶囊对乳腺增生大鼠血清雌二醇(E_(2))、孕酮(P)、泌乳素(PRL)、促黄体生成素(LH)、促卵泡激素(FSH)及乳腺组织中雌激素受体(ER)、孕激素受体(PR)、P53的影响。方法70只SPF级雌性未孕SD大鼠,随机分空白组10只、造模组60只。造模组60只大鼠雌孕激素联合造成乳腺增生模型,分为模型组、乳癖消组、他莫昔芬组及慈莲胶囊低、中、高剂量组。干预30 d后,用酶联免疫法(ELISA)测血清中E_(2)、P、PRL、LH、FSH含量。免疫组化SP法测乳腺组织中ER、PR、P53。结果他莫昔芬组、乳癖消组、慈莲胶囊各组均能不同程度降低血清E_(2)、PRL、FSH含量(P<0.05),他莫昔芬组、乳癖消组能升高P的水平(P<0.05)。慈莲胶囊各组均能升高P含量,但差异无统计学意义。他莫昔芬组、乳癖消组、慈莲胶囊各组均可升高LH的水平(P<0.05)。与模型组相比,他莫昔芬组、乳癖消组、慈莲胶囊各组ER、PR、P53表达均降低(P<0.05)。结论慈莲胶囊对乳腺增生大鼠的治疗作用可能与其可以调节血清性激素水平及乳腺组织中ER、PR和P53有关。

Objective To investigate the effects of Cilian capsule on serum estradiol(E_(2)), progesterone(P), prolactin(PRL), luteinizing hormone(LH), follicle-stimulating hormone(FSH), estrogen receptor(ER), progesterone receptor(PR) and P53 in mammary gland hyperplasia rats. Methods Seventy SPF female non-pregnant SD rats were randomly divided into blank group(n = 10) and model group(n= 60). A total of 60 rats in the model group were divided into model group, Rupixiao group, tamoxifen group and Cilian capsule low-dose, medium-dose and high-dose groups. After 30 days of intervention, the contents of E_(2), P, PRL, LH and FSH in serum were measured by enzyme-linked immunosorbent assay(ELISA). Immunohistochemical SP method was used to detect ER, PR and P53 in breast tissue. Results The contents of serum E_(2), PRL and FSH in the tamoxifen group, Rupixiao group and Cilian capsule groups were decreased to different degrees(P<0.05), and the levels of P in the tamoxifen group and Rupixiao group were increased(P<0.05). Cilian capsule could increase P content in all groups, but the difference was not statistically significant. tamoxifen group, Rupixiao group and Cilian capsule groups could increase the level of LH(P<0.05). Compared with the model group, the expressions of ER, PR and P53 in tamoxifen group, Rupixiao group and Cilian capsule groups were decreased(P<0.05). Conclusion The therapeutic effect of Cilian capsule on rats with mammary hyperplasia may be related to the regulation of serum sex hormone levels and the levels of ER, PR and P53in mammary tissue.
