摘要
胶体金免疫层析(GICA)是目前唯一能用于现场检测微囊藻毒素(MC)-LR的技术。现场使用GICA存在藻细胞内MC-LR无法检测、检出限随环境温度变化、基质效应等问题。为方便水源现场MC-LR总量(TMC-LR)的调查和预警,开发了“煮沸—浓缩—PES膜过滤—调p H(可选)”前处理方法并优化了相关参数。结果表明,检测卡能耐受2~12的p H、2 g/L的蛋白和200 g/L的多糖。该方法的最佳煮沸时间是30 min,浓缩1倍能保证在10~37℃环境温度下1μg/L的检出限。用卵囊藻模拟进入藻华预警阶段的水体,采用模拟样品和实际样品进行了方法技术性能确认,结果满足筛选检测要求。该方法可用于现场快检地表水中的TMC-LR。
Gold immunochromatography colloidal assay(GICA) is the only technique available to detect microcystin(MC)-LR on-site. However, there are some problems in field application of GICA, such as the inability to detect MC-LR in algal cells, the change of detection limit with ambient temperature and matrix effect. To facilitate the on-site investigation and early warning of the total MC-LR(TMC-LR) at water sources, the pretreatment method of boiling, concentration, PES membrane filtration and pH adjustment(optional) was developed and the related parameters were optimized. The limits of pH, protein and polysaccharide tolerated by the test card were 2-12, 2 g/L and 200 g/L, respectively. The optimum boiling time was 30 min, and the detection limit of 1 μg/L at ambient temperature of 10-37 ℃ could be ensured by 1 time concentration. Oocystis was used to simulate the early warning stage of algal bloom in a water body, the technical performance of the method was verified by using simulated and real water samples, and the results met the requirements of selection and detection, indicating that the method could be used for rapid detection of TMC-LR in surface water.
