L-SeMC对酒精致中枢兴奋小鼠学习记忆和自发活动的影响

摘要 目的探讨L-硒-甲基硒代半胱氨酸(L-selenium-methylselenocysteine,L-SeMC)对酒精致中枢兴奋小鼠学习记忆和自发活动的影响。方法将雄性昆明小鼠随机分为对照组、酒精组及L-SeMC低、中和高剂量组,分别灌胃给予L-SeMC 10.4、13.0、39.0μg·kg^(-1),30 min后给予酒精1.5 g·kg^(-1),进行自发活动检测、跳台实验和避暗实验,检测血清ALT和AST、肝脏组织MDA含量及SOD活力。结果10.4、13.0、39.0μg·kg^(-1)L-SeMC可显著减少酒精致中枢兴奋小鼠活动次数和站立次数;10.4μg·kg^(-1)L-SeMC可显著延长跳台实验潜伏期,13.0μg·kg^(-1)L-SeMC可显著延长潜伏期,减少错误次数;10.4μg·kg^(-1)L-SeMC可显著延长避暗实验潜伏期,减少错误次数。3个剂量L-SeMC可降低血清ALT、AST水平,降低肝脏组织MDA含量,增强SOD活性。结论10.4、13.0μg·kg^(-1)L-SeMC可改善酒精致中枢兴奋对小鼠学习记忆功能的降低,抑制酒精引起的中枢兴奋自发活动,其机制可能与改善酒精致中枢兴奋小鼠的肝功能和肝脏氧化损伤有关。 Objective To investigate the effects of L-Se-methylselenocysteine(L-SeMC)on learning memory and spontaneous activity in alcohol-induced central excitation mice.Methods Male kunming mice were randomly divided into control group,alcohol group and low,medium and high dose L-SeMC groups.The mice were given 10.4,13.0 and 39.0μg·kg^(-1)L-SeMC by intragastric administration,respectively.After 30 minutes,the mice were administrated 1.5 g·kg^(-1)alcohol for spontaneous activity detection,step-down test and dark avoidance test,ALT,AST,MDA and SOD measurement.Results 10.4,13.0 and 39.0μg·kg^(-1)L-SeMC could significantly reduce the activity times and standing times of alcohol-induced central excitation mice.10.4μg·kg^(-1)L-SeMC obviously prolonged the latency of step-down test,13.0μg·kg^(-1)L-SeMC remarkably prolonged the latency and reduced the number of errors.10.4μg·kg^(-1)L-SeMC significantly prolonged the latency and reduced the number of errors of dark avoidance test.The 3 doses of L-SeMC could decrease the levels of ALT,AST,MDA,and increase the activity of SOD.Conclusion 10.4,13.0μg·kg^(-1)L-SeMC could ameliorate the decrease of learning memory function and inhibit the spontaneous activity in alcohol-induced central excitation mice,which might be related to the improvement of liver function and liver oxidative damage.
