白藜芦醇对白内障小鼠晶状体透明度的作用

摘要 目的探讨白藜芦醇对白内障小鼠晶状体透明度的改善作用及对上皮细胞凋亡的影响,并阐明其作用机制。方法用紫外线照射建立白内障小鼠模型,RT-qPCR检测建模与未建模小鼠晶状体miR-125b表达水平;建模小鼠随机分为模型组、白藜芦醇组和白藜芦醇+inhibitor组,另设对照组,相应干预后,比较各组小鼠晶状体浑浊度及评分;检测晶状体超氧化物歧化酶(SOD)、谷胱甘肽(GSH)和丙二醛(MDA)水平;流式细胞术检测晶状体上皮细胞(LEC)凋亡率;RT-qPCR和Western blot分别检测晶状体p53、B细胞淋巴瘤/白血病-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)mRNA和蛋白表达。结果造模后小鼠晶状体miR-125b表达水平明显低于未造模小鼠(P<0.05)。白藜芦醇可明显降低白内障小鼠晶状体浑浊度及评分,提高晶状体SOD、GSH水平,降低MDA水平及LEC凋亡率,降低p53、Bax mRNA和蛋白表达水平,升高Bcl-2 mRNA和蛋白表达水平。结论白藜芦醇可改善白内障小鼠晶状体浑浊度,减少细胞凋亡,可能与促进miR-125b表达有关。 Objective To investigate the effect of resveratrol on improving lens transparency in cataract mice and its effect on epithelial cell apoptosis,and to clarify its possible mechanism.Methods Ultraviolet radiation was used to establish a mouse model of cataract.RT-qPCR was used to detect the expression level of miR-125 b in the lens of modeled and unmodeled mice.The modeled mice were randomly divided into model group,resveratrol group,resveratrol+inhibitor group,and control group.After corresponding intervention,the lens turbidity and score,lens superoxide dismutase(SOD),glutathione(GSH),and malondialdehyde(MDA)levels were detected and compared.Flow cytometry was used to detect the apoptotic rate of lens epithelial cells(LEC).RT-qPCR and Western blotting were used to detect lens p53,B-cell lymphoma/leukemia-2 gene(Bcl-2),Bcl-2 associated X protein(Bax)mRNA and protein expression.Results The expression level of miR-125 b in the lens of modeled mice was significantly lower than that of unmodeled mice(P<0.05).Resveratrol can significantly reduce lens turbidity and score of cataract mice,increase lens SOD and GSH levels,reduce MDA levels and LEC apoptosis rate,reduce p53,Bax mRNA and protein expression levels,and increase Bcl-2 mRNA and protein expression.Conclusion Resveratrol can improve lens turbidity and reduce cell apoptosis in mice with cataract,which may be related to the promotion of miR-125 b expression.
