当归提取物对卵巢功能不全大鼠卵巢功能的作用 被引量:1

摘要 目的探讨当归提取物对早发性卵巢功能不全(POI)大鼠卵巢功能的改善是否与Nrf2/HO-1通路介导的炎性调节有关。方法取60只SD雌性大鼠,其中48只通过雷公藤多苷片建立POI模型,造模成功后随机分为模型组及当归提取物低、高剂量组和西药组,其余12只为对照组。当归提取物低、高剂量组分别给予当归提取物25、100 mg·kg^(-1),连续灌胃15 d,西药组给予戊酸雌二醇0.12 mg·kg^(-1),连续灌胃15 d,对照组及模型组给予等量生理盐水,灌胃15 d。测定血清中E_(2)、AMH、LH和FSH的水平;HE染色观察各组大鼠卵巢组织的病理变化和卵泡计数;用RT-qPCR和Western blot检测卵巢组织中Nrf 2和HO-1 mRNA和蛋白的表达情况。结果与模型组比较,其余3组血清E_(2),AMH,原始卵泡、窦前卵泡、窦状卵泡的数量,Nrf 2、HO-1 mRNA和蛋白的表达水平增加,FSH、LH、TNF-α、IL-4、IL-6水平和闭锁卵泡数量减少(P<0.05)。与当归提取物低剂量组比较,当归提取物高剂量组和西药组血清E_(2)、AMH、卵泡数量、Nrf 2、HO-1 mRNA和蛋白的表达水平增加,FSH、LH、TNF-α、IL-4、IL-6水平和闭锁卵泡数量减少(P<0.05)。当归提取物高剂量组与西药组上述指标比较,差异无统计学意义。结论当归提取物可通过激活Nrf 2/HO-1信号通路改善炎性因子水平,在一定程度上修复POI大鼠的卵巢功能,发挥卵巢保护作用。 Objective To investigate whether the improvement of Angelica sinensis extract(ASE)on ovarian function in rats with early-onset ovarian insufficiency(POI)is related to the inflammatory regulation mediated by NRF 2/HO-1 pathway.Methods 60 female SD rats were selected,of which 48 were established POI model by Tripterygium Wilfordii Polyglycoside Tablets.After successful modeling,they were randomly divided into model group,low and high dose groups and western medicine group,and the other 12 were grouped as the control.The low and high dose group were given 25 and 100 mg·kg^(-1)ASE,the western medicine group was given 0.12 mg·kg^(-1)estradiol valerate,and the control group and model group were given the same amount of normal saline for 15 days.The levels of serum E_(2),ABH,LH,FSH and follicle count were measured.HE staining was used to observe the changes in ovarian tissue and follicle count.RT-PCR and Western blot were used to detect the protein and mRNA expression of NRF 2 and HO-1 in ovarian tissues.Results Compared with the model group,the levels of serum E_(2),AMH,primordial follicles,preantral follicles,the numbers of sinus follicles,Nrf 2,HO-1 mRNA and protein expression were increased,FSH,LH and TNF-α,IL-4,IL-6 and the number of atretic follicles were decreased in the low and high dose groups and western medicine group(P<0.05).Compared with the low dose group,the levels of serum E_(2),AMH,primordial follicles,preantral follicles,number of sinus follicles,Nrf 2,HO-1 mRNA and proteinexpression were increased,FSH,LH and TNF-α,IL-4,IL-6 and the numbers of atretic follicles were decreased in the high dose group and western medicine group(P<0.05).There was no significant difference in the above indexes between the high dose of group and the western medicine group.Conclusion ASE can improve the level of inflammatory factors by activating Nrf2/HO-1 signal pathway,repair the ovarian function of POI rats to a certain extent and play an ovarian protective role.
