艾叶挥发油对H_(2)O_(2)诱导鲤鱼肝脏氧化损伤的保护作用 被引量:7

摘要 为探讨艾叶挥发油对H_(2)O_(2)诱导的鲤鱼肝脏氧化损伤的保护作用,本试验以H_(2)O_(2)为诱导剂构建鲤鱼肝脏氧化应激损伤模型,分别投喂添加0.1、0.2和0.4 g·kg^(-1)艾叶挥发油的饵料对其进行保护。试验进行30 d后,采集鲤鱼的血清和肝脏,检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)活性和总蛋白(TP)、白蛋白(ALB)、丙二醛(MDA)含量;采用组织学方法对鲤鱼肝组织切片进行显微观察;使用实时荧光定量PCR(qRT-PCR)技术检测Cu/Zn-SOD、Mn-SOD、GPx、CAT和核转录因子(Nrf2)mRNA的相对表达量。结果表明,H_(2)O_(2)暴露30 d后,鲤鱼血清中ALT、AST活性升高,TP、ALB含量下降;血清和肝脏中SOD、CAT、GPx活性降低,MDA含量上升,H_(2)O_(2)导致肝脏组织病理损伤,抗氧化基因表达受到抑制。而艾叶挥发油能够抑制ALT、AST活性的上升和TP、ALB含量的下降,提高机体的抗氧化能力,缓解肝脏组织的病理损伤,激活Nrf2诱导细胞抗氧化基因的表达。综上所述,艾叶挥发油对H_(2)O_(2)诱导的鲤鱼肝脏氧化损伤具有一定的保护作用。本研究可为进一步探究艾叶挥发油作为绿色安全的抗氧化剂提供思路和基础资料。 In order to explore the protective effect of Artemisia argyi essential oils(AAEO)on H_(2)O_(2)-induced oxidative damage of carp liver,the oxidative injury model of carp liver was established using H_(2)O_(2) as an inducer,and the diets containing 0.1,0.2,and 0.4 g·kg^(-1) AAEO were fed to protect it.After treatment for 30 days,the serum and liver were collected to detect the activities of alanine aminotransferase(ALT),aspartate aminotransferase(AST),superoxide dismutase(SOD),catalase(CAT),glutathione peroxidases(GPx),and the contents of total protein(TP),albumin(ALB),and malondialdehyde(MDA).The structure of liver tissues in carp was observed by histological method.The relative expression levels of Cu/Zn SOD,Mn-SOD,GPx,CAT,and transcription factor(Nrf2)were detected by quantitative real-time PCR.The results showed that the exposure of H_(2)O_(2) for 30 days could increase the activities of ALT and AST,while decrease the contents of TP and ALB in serum,H_(2)O_(2) could reduce the activities of SOD,CAT,and GPx,while elevate the MDA concentration in serum and liver.H_(2)O_(2) could cause the pathological damage and inhibit the expression of antioxidant genes of liver tissues.AAEO could inhibit the increase of ALT and AST activities and the decrease of TP and ALB contents,improve the antioxidant capacity of the body,relieve the pathological damage of liver tissue,and induce the expression of antioxidant genes by activating Nrf2.In conclusion,AAEO has a certain protective effect on H_(2)O_(2)-induced oxidative damage of carp liver,which can provide basic information for further exploring AAEO as a green and safe antioxidant.
