摘要 研究了Rhizopussp .PW35 8菌的固态生长和产脂肪酶条件。结果表明 :黄豆饼粉为培养基的基本成分 ,用来生产脂肪酶。培养基中可加入淀粉和蛋白胨作为碳源和氮源 ,有利于脂肪酶的合成。培养基的含水量以及金属离子Ca2 +、Mg2 +的浓度也影响Rhizopussp .PW35 8菌的脂肪酶产生。在优化条件下 ,12g豆饼粉中含 1 0g淀粉及 0 .5g蛋白胨、15ml营养盐中Ca2 +、Mg2 +离子浓度分别为 8.0和 4.0g/L ,培养基含水量为 5 5 .6 % ,在接种后培养 48h ,酶活力可达最大值 32 0IU/g干培养基。脂肪酶的基本性质研究表明 ,酶的最适反应温度和pH分别为 35℃和 7 0 ,酶的半失活温度为 5 3.5℃ ,不同的pH环境中 ,30℃保温1h后酶在pH6 .5~ 8. Growth and enzyme production of Rhizopus sp.PW358 by solid state fermentation were studied.The result showed that the soybean cake could be used as component of medium.Starch and peptone used as carbon and nitrogen sources benefited for the lipase production.The water content and the concentration of metal ions such as Ca 2+and Mg 2+in the medium effected on the enzyme production.Under optimal condition,starch 1.0g,peptone 0.5g,soybean cake 12g,the concentration of Ca 2+ and Mg 2+ 8.0 and 4.0 g/L respectively,the water content of medium 55.6%,lipase activity reached 320 IU/g dry medium after cultured 48h at 28℃. The properties of lipase were also studied.The optimal temperature and pH were 35℃ and 7.0 respectively.The half lost temperature were 53.5℃.Lipase was relative stable at pH range from 6.5 to 8.5 after incubated in different pH solution at 30℃.
