桥本氏甲状腺炎和乳头状甲状腺癌核心蛋白的筛选及临床意义 被引量:3

摘要 目的 分析甲状腺炎与乳头状甲状腺癌核心基因的关系,为分析桥本氏甲状腺炎发展和诊断乳头状甲状腺癌提供新方法。方法 预测桥本氏甲状腺炎与乳头状甲状腺癌的共同基因。采用Cyto Hubba 0.1筛选核心基因编码的蛋白质。采用Metascape对核心蛋白生物学过程和KEGG通路进行富集分析。结果 桥本氏甲状腺炎与乳头状甲状腺癌共同基因有147个,核心蛋白包括白介素(IL-6)、肿瘤蛋白(TP53)、血管内皮生长因子A(VEGFA)、肿瘤坏死因子(TNF)、胰岛素(INS)。核心蛋白参与细胞增殖相关的生物过程包括调控细胞增殖、单核细胞增殖、T细胞增殖等46条;组织重塑、血管生成与发育、血管内皮细胞迁移等22条生物学过程与血管生长有关。KEGG通路包括甲状腺激素信号通路、甲状腺癌等15条通路。结论核心蛋白对分析桥本氏甲状腺炎和乳头状甲状腺癌关系,判断桥本氏甲状腺炎演变和诊断乳头状甲状腺癌具有重要价值。 Objective To reveal the core gene relationship between thyroiditis and papillary thyroid carcinoma, and to propose a new method for analyzing the development of Hashimoto’s thyroiditis and diagnosing papillary thyroid carcinoma. Methods The common genes of Hashimoto’s thyroiditis and papillary thyroid carcinoma were predicted. Cyto Hubba 0.1 was used to screen the protein encoded by the core gene. Metascape was applied to perform enrichment analysis on core protein biological process and KEGG pathways. Results There were 147 common genes between Hashimoto’s thyroiditis and papillary thyroid carcinoma. The core proteins included interleukin(IL-6), tumor protein(TP53), vascular endothelial growth factor A(VEGFA), tumor necrosis factor(TNF) and insulin(INS). Core proteins were involved in 46 biological processes related to cell proliferation, including regulating cell proliferation, monocyte proliferation and T cell proliferation. 22 biological processes such as tissue remodeling, angiogenesis and development, and migration of vascular endothelial cells were related to vascular growth. 15 KEGG pathways including thyroid hormone signaling pathway and thyroid carcinoma were involved. Conclusion Core protein has important value in analyzing the relationship between Hashimoto’s thyroiditis and papillary thyroid carcinoma, judging the evolution of Hashimoto’s thyroiditis and diagnosing papillary thyroid carcinoma.
