加味黑布膏对大鼠耳廓复合痤疮模型外周血清IL-1α、IL-4水平的影响

Influences of modified Heibu ointment on IL-1α and IL-4 levels in peripheral blood of rats with auricle compound acne model

目的观察加味黑布膏对Wistar大鼠耳廓复合痤疮模型皮损程度及外周血清白细胞介素-1α(IL-1α)、白细胞介素-4(IL-4)水平的影响。方法将62只雌雄各半的Wistar大鼠随机分为空白组、模型组及4个药物组(选取2只大鼠判断耳廓复合痤疮模型形成),空白组、模型组不使用药物干预,其余四组予相应药物涂抹,其中阳性对照组外用维A酸乳膏,加味黑布膏低、中、高剂量组外用不等剂量的加味黑布膏。4周后肉眼观察各组鼠耳皮损程度;比较各组大鼠血清IL-1α、IL-4水平。结果涂药4周后,与模型组比较,药物组鼠耳皮损均有不同程度地改善,其中阳性对照组、高剂量组皮损症状基本消退。阳性对照组、加味黑布膏各剂量组的IL-1α、IL-4水平显著低于模型组(P<0.05);高剂量组的IL-1α、IL-4水平显著低于中、低剂量组(P<0.05)。结论加味黑布膏可明显改善Wistar大鼠耳廓复合痤疮模型处皮损症状,可能与降低外周血清IL-1α、IL-4水平有关。

Objective To observe the influences of modified Heibuo intment on the skin lesion degree of Wistar rats with auricle compound acne model and interleukin-1α(IL-1α)and interleukin-4(IL-4)levels in peripheral blood.Methods Sixty-two Wistar rats,half male and half female,were randomly divided into blank group,model group and 4drug groups(2 rats were selected to determine the formation of auricle compound acne model).No drug intervention was used in the blank group and the model group,and the other 4 groups were applied with corresponding drugs,among which the positive control group was treated with tretinoin cream for external use,and the low-dose,medium-dose and high-dose groups were treated with different doses of modified Heibu ointment for external use.The degree of mouse ear skin lesion in each group was observed by naked eye after 4 weeks;the levels of serum IL-1αand IL-4 in each group were compared.Results After 4 weeks of drug application,compared with the model group,the mouse ear skin lesion in the drug group improved to varying degrees,and the symptoms of skin lesion in the positive control group and the high-dose group basically subsided.The levels of IL-1αand IL-4 in the positive control group and the each dose group of modified Heibu ointment were significantly lower than those in the model group(P<0.05);the levels of IL-1αand IL-4 in the high-dose group were significantly lower than those in the medium-dose and low-dose group(P<0.05).Conclusion Modified Heibu ointment can obviously improve the symptoms of skin lesion in the Wistar rats with auricle compound acne model,which may be related to the reduction of serum IL-1αand IL-4 levels in peripheral blood.
