子痫前期孕妇胎盘组织中 BRCA1、MIC-1的表达情况及临床意义 被引量:1

Expressions and clinical significance of BRCA1 and MIC-1 in the placental tissues of pregnant women with preeclampsia

摘要 目的探讨子痫前期孕妇胎盘组织中乳腺癌Ⅰ型易感基因(BRCA1)、巨噬细胞抑制因子-1(MIC-1)的表达情况及临床意义。方法选取经剖宫产分娩的80例子痫前期孕妇,将其分为轻度子痫前期组40例与重度子痫前期组40例;选取同期经剖宫产分娩的40例正常妊娠孕妇为对照组。收集3组研究对象的临床资料,并检测胎盘组织中BRCA1和MIC-1的mRNA和蛋白表达水平。分析子痫前期孕妇胎盘组织中BRCA1与MIC-1的mRNA表达水平之间的相关性,以及两者与患者血压、24 h尿蛋白水平、血脂水平的相关性。结果对照组、轻度子痫前期组、重度子痫前期组孕妇收缩压、舒张压、平均动脉压、24 h尿蛋白及LDL-C水平依次升高,而HDL-C水平依次降低,且轻度子痫前期组和重度子痫前期组孕妇的总胆固醇水平均高于对照组(均P<0.05)。对照组、轻度子痫前期组、重度子痫前期组孕妇胎盘组织中BRCA1蛋白阳性表达及mRNA表达水平依次降低,而MIC-1蛋白阳性表达及mRNA表达水平依次增加(均P<0.05)。子痫前期孕妇胎盘组织中BRCA1与MIC-1的mRNA表达水平呈负相关(P<0.05);子痫前期孕妇胎盘组织中BRCA1的mRNA表达水平与收缩压、舒张压、平均动脉压、24 h尿蛋白水平、总胆固醇水平、LDL-C水平呈负相关,与HDL-C水平呈正相关,而MIC-1的mRNA表达水平与上述指标的相关性相反(均P<0.05)。结论子痫前期孕妇胎盘组织中BRCA1呈低表达而MIC-1呈高表达,两者可能共同参与子痫前期的发生。 Objective To investigate the expressions and clinical significance of breast cancer typeⅠsusceptibility gene(BRCA1)and macrophage inhibitory cytokine-1(MIC-1)in placental tissues of pregnant women with preeclampsia.Methods A total of 80 pregnant women with preeclampsia undergoing Cesarean section delivery were selected,and they were assigned to mild preeclampsia group(40 cases)or severe preeclampsia group(40 cases),and 40 normal pregnant women undergoing Cesarean section delivery during the same period were selected as control group.The clinical data of the research subjects in the three groups were collected.The mRNA and protein expressions of BRCA1 and MIC-1 in placental tissues were detected.The correlations of BRCA1 mRNA expression with MIC-1 mRNA expression,as well as the two as above with patients′blood pressures,24-hour urine protein level,and blood lipid level were analyzed.Results The pregnant women in the control group,mild and severe preeclampsia groups exhibited successively elevated systolic pressure,diastolic pressure,mean arterial pressure,and successively elevated levels of 24-hour urine protein and LDL-C,whereas a successively decreased level of HDL-C,and the level of total cholesterol was higher in the mild and severe preeclampsia groups than in the control group(all P<0.05).The pregnant women in the control group,and mild and severe preeclampsia groups acquired successively decreased protein positive expression and mRNA expression of BRCA1,whereas successively increased protein positive expression and mRNA expression of MIC-1 in placental tissues(all P<0.05).The mRNA expressions of BRCA1 and MIC-1 in placental tissues of pregnant women with preeclampsia were negatively correlated(P<0.05);moreover,mRNA expression of BRCA1 in placental tissues of pregnant women with preeclampsia negatively correlated with systolic pressure,diastolic pressure,mean arterial pressure,24-hour urine protein level,total cholesterol level and LDL-C level,and positively correlated with HDL-C level,whereas the correlation of mRNA expression of MIC-1 with the aforesaid indices was contrary(all P<0.05).Conclusion BRCA1 in placental tissues of pregnant women with preeclampsia presented as low-expression,while MIC-1 as high-expression,and the two may be jointly involved in the occurrence of preeclampsia.
