菟丝子-桑寄生改善小鼠卵巢储备功能及子宫内膜容受性 被引量:5

摘要 目的:探究菟丝子、桑寄生对小鼠卵巢储备功能及子宫内膜容受性的调节作用及机制。方法:使用羟基脲联合米非司酮构建小鼠子宫内膜容受性障碍模型,使用环磷酰胺制备小鼠卵巢储备功能下降模型,采用实时荧光定量PCR(qPCR)及酶联免疫吸附测定(ELISA)等方法检测菟丝子、桑寄生单药及菟丝子-桑寄生配伍对小鼠子宫组织中相关基因表达及血清中激素水平的影响。结果:菟丝子-桑寄生显著改善子宫内膜容受性障碍模型小鼠MMP-9/TIMP-3表达平衡,上调LIF及EGF表达(p<0.05);改善卵巢储备功能下降模型小鼠血清中E2、LH、FSH、AMH激素水平。结论:菟丝子-桑寄生能够通过调节MMP-9/TIMP-3表达平衡改善小鼠子宫内膜容受性障碍,通过调节模型小鼠相关激素水平,改善卵巢功能。 Objective: To explore the regulatory effect and mechanism of Tusizi and Sangjisheng on ovarian reserve function and endometrial receptivity in mice. Method: The endometrial receptive disorder model was established by using hydroxyurea and mifepristone. The ovarian reserve function decline model was prepared by using cyclophosphamide. Real-time fluorescence quantitative PCR(qPCR) and enzyme-linked immunosorbent assay(ELISA) assays were performed to evaluate the effects of Tusizi and Sangjisheng on the expression of related genes in mice uterus and the level of hormones in serum. Result: Tusizi-Sangjisheng significantly improved the expression balance of MMP-9/TIMP-3 and up-regulated the expression of LIF and EGF in mice with endometrial receptivity disorder(p < 0.05), improved the levels of E2, LH, FSH and AMH hormones in serum of mice with decreased ovarian reserve function. Conclusion: Tusizi-Sangjisheng can improve endometrial receptivity disorder in mice by regulating the expression balance of MMP-9/TIMP-3, and improve ovarian function by regulating the level of related hormones in model mice.
