HPLC法同时测定补益中药中尿苷、腺嘌呤、鸟苷和腺苷含量分析 被引量:2

摘要 目的应用高效液相色谱(high performance liquid chromatography,HPLC)法分析6种常用补益中药(熟地黄、肉苁蓉、黄芪、人参、麦冬、当归)中腺嘌呤、尿苷、腺苷及鸟苷的含量。方法连云港市中医院2021年1月采用HPLC法同时测定补益中药中各成分含量,分析柱选取Zorbax SB-Aq型,参数为5μm、250.0 mm×4.6 mm,以乙腈+0.05%甲酸水溶液作为流动相梯度,洗脱处理,流速控制在0.5 mL/min,设置检测波长为260nm,柱温设置为30℃。结果在0.30~77.32μg/mL、0.35~90.73μg/mL、0.23~58.29μg/mL及0.61~157.07μg/mL范围内,腺嘌呤、尿苷、腺苷及鸟苷的线性关系均良好(r=1.000、1.000、0.999、1.000),加样回收率(n=3)分别为103.4%(RSD=1.7%)、98.3%(RSD=0.6%)、97.2%(RSD=1.8%)及97.9%(RSD=0.8%);当归、人参、熟地黄、肉苁蓉、黄芪及麦冬中均含有核苷类成分,其中黄芪、人参及当归中均含有腺嘌呤、尿苷、腺苷及鸟苷成分,且具有较高含量。结论在补益中药中核苷类成分可能是一类重要的活性成分。 Objective To determine the content of adenine,uridine,adenosine and guanosine in 6 kinds of tonic traditional Chinese medicines(Radix Rehmanniae Preparata,herba Cistanches,radix Astragali,radix Ginseng,Radix Ophiopogonis and Angelicae sinensis)by high performance liquid chromatography(HPLC).Methods HPLC method was used to determine the content of each component in tonic traditional Chinese medicine in Lianyungang Hospital of Traditional Chinese Medicine in January 2021.Zorbax SB-Aq analytical column was as the analytical column with the parameters of 5μm and 250.0 mm×4.6 mm.The mobile phase consisted of acetonitrile and+0.05%formic acid.The mobile phase was gradient elution with the flow rate of 0.5 mL/min.The detection wavelength was set at 260 nm and the column temperature was set at 30℃.Results The linear ranges of adenine,uridine,adenosine and guanosine were 0.30-77.32μg/mL,0.35-90.73μg/mL,0.23-58.29μg/mL and 0.61-157.07μg/mL,respectively(r=1.000,1.000,0.999,1.000).The average recoveries(n=3)were 103.4%(RSD=1.7%),98.3%(RSD=0.6%),97.2%(RSD=1.8%)and 97.9%(RSD=0.8%),Angelica sinensis,radix Ginseng.Radix Rehmanniae Preparata,herba Cistanches,radix Astragali and Radix Ophiopogonis all contain nucleosides,among which radix Astragali,radix Ginseng and Angelicae sinensis all contain adenine,uridine,adenosine and guanosine,and have a higher content.Conclusion Nucleosides in tonic Chinese medicine may be an important active ingredient.
