P(NIPAAm-co-AM)智能水凝胶的制备及其药物缓释作用 被引量:1

摘要 采用水相引发氧化还原体系聚合方法制备比表面积大、孔容大与孔道存在交联互穿结构的温敏性水凝胶(P(NIPAAm-co-AM),PNA).利用傅里叶红外光谱、电子扫描显微镜和氮气吸附—脱附实验对制备的3种水凝胶进行了分析与结构表征,同时还系统地研究了水凝胶的力学性能和溶胀动力学性能,以及温度变化对水凝胶溶胀性能的影响,探索出了制备性能优异的智能水凝胶的配比,揭示了单体配比对智能水凝胶结构与性能的影响.此外,选择多肽谷胱甘肽为包载药物做了体外药物包载—释放实验,进一步明确了水凝胶结构与药物的包载和释放之间的关系,对于实现药物分子体内的包载与释放具有重要意义. The temperature-sensitive hydrogels(P(NIPAAm-co-AM),PNA)with large specific surface area,large pore volume and cross-linking interpenetrating structure were prepared by using aqueous redox system as initiator.The property and structure of a series of prepared PNA hydrogels were analyzed and characterized by Fourier-Transform Infrared Spectroscopy,Scanning Electron Microscopy and nitrogen adsorption.Meanwhile,the swelling kinetic properties and mechanical properties of PNA hydrogels and the effects of the temperature changes on the swelling properties were investigated in details.The ratio for the preparation of the high-quality hydrogel were explored.The results showed that the influence of monomer ratios on the structure and properties of intelligent hydrogel were revealed.In addition,in vitro drug release were performed by using the polypeptide glutathione as slow-release drug,which further clarified the relationship between hydrogel structure and encapsulation and release of the drug.This research has important scientific significance and wide application prospect for drug encapsulation and release in vivo.
