中药复方液对麻黄素致仔鼠脑组织氧化损伤的影响

摘要 为研究中草药对麻黄素致仔鼠脑组织氧化损伤的影响,用递增剂量(2.0,3.0,4.0 g·L^(-1))给72只仔鼠连续腹腔注射麻黄素溶液15 d,每次注射麻黄素溶液后,再灌胃中药复方液(2.0 g·L^(-1))进行干预,在显微镜下分析研究额叶皮层结构的变化,用比色法检测分析脑组织SOD和CAT活性以及MDA含量的变化,用免疫组织化学法检测分析脑组织Bax蛋白表达的变化.结果显示,模型组仔鼠脑组织SOD,CAT活性均低于对照组,MDA含量高于对照组,差异显著(P<0.05)或极显著(P<0.01);模型组额叶皮层组织有不同程度的损伤,细胞数目减少,形态结构出现改变;模型组脑组织阳性表达的Bax蛋白大于对照组,差异极显著(P<0.01).与模型组比较,中药组脑组织SOD,CAT活性又升高,MDA含量降低,差异显著(P<0.05)或极显著(P<0.01),额叶脑细胞结构明显改善,Bax蛋白的阳性表达密度降低.上述结果表明,中药复方液可提高细胞的抗氧化能力,抑制脑组织Bax蛋白的表达量,能够减轻或减缓麻黄素兴奋剂对脑结构的损伤. To research the effect of Chinese medicine compound on ephedrine-induced oxidative damage in mouse brain,72 filial mice were intraperitoneally injected escalation with doses(2.0,3.0,4.0 g·L^(-1))ephedrine 0.2 mL continuous(1~5 d,6~10 d and 11~15 d)for 15 d(twice a day).After injection ephedrine 1 h,the mice were given a gavage of Chinese medicine for withdrawal respectively.The frontal lobe of tissue structure were observed by biomicroscopy,the brain SOD,CAT activity and MDA content were detected by colorimetry,the changes of Bax protein expression in brain was detected by immunohistochemistry method.The results showed that the SOD and CAT activity of ephedrine group in brain are decreased significantly,MDA content are increased significantly compared with the control group(P<0.05 or P<0.01).The structure of frontal lobe of ephedrine group is damaged in varying degree,and cell number is reduced.The expression of Bax protein in brain is increased,and the average optical density is increased significantly(P<0.01).Compared with the ephedrine group,the SOD and CAT activity of Chinese medicine group in brain are increased significantly,and MDA content is decreased significantly(P<0.05 or P<0.01).The situation is improved in the Chinese medicine group,the expression of Bax protein in brain is decreased.All results indicated that Chinese medicine can improve the anti-oxidation ability of cells and inhibit the expression of Bax protein in brain.It can effectively reduce the damage of ephedrine on the brain and has a certain protective effect on the brain.
