摘要
目的观察子宫腺肌症患者血清中沉默调节蛋白6(Sirt6)、氧化应激指标(丙二醛、超氧化物歧化酶)的水平,并分析其相关性。方法收集2021年4—10月同济大学附属杨浦医院收治的子宫腺肌症患者20例作为腺肌症组,另选择同期体检健康妇女20名作为正常组。酶联免疫吸附法检测血清Sirt6水平;试剂盒检测丙二醛含量和超氧化物歧化酶活性。采用Pearson相关分析法评估子宫腺肌症患者血清中Sirt6水平与丙二醛含量和超氧化物歧化酶活性之间的关系。结果正常组血清中Sirt6水平明显高于腺肌症组[(7.03±4.59)ng·mL^(-1)比(2.65±1.52)ng·mL^(-1),P<0.001],丙二醛含量明显低于腺肌症组[(2.75±2.11)μmol·L^(-1)比(5.45±2.48)μmol·L^(-1),P<0.001],而超氧化物歧化酶活性高于腺肌症组[(0.75±0.20)U比(0.52±0.24)U,P<0.01];子宫腺肌症患者血清中Sirt6水平与超氧化物歧化酶活性呈正相关(r=0.33,P<0.05),而与丙二醛含量呈负相关(r=-0.56,P<0.05)。结论子宫腺肌症患者血清中Sirt6和氧化应激水平均升高,且Sirt6水平与氧化应激水平相关,提示Sirt6可能参与子宫腺肌症氧化损伤的调控机制。
Objective To observe the serum levels of silencing information regulatory protein 6(Sirt6)and oxidative stress indicators,ie.malondialdehyde(MDA)and superoxide dismutase(SOD)in patients with adenomyosis,and to analyze their correlations.Methods Twenty patients with adenomyosis admitted to Yangpu Hospital Affiliated to Tongji University from April to October 2021 were collected as the adenomyosis group,and 20 healthy women were chosen in the same period as the normal group.Serum Sirt6 levels were detected by enzyme-linked immunosorbent assay.The content of MDA and the activity of SOD were measured using the reagent kits.Pearson correlation analysis was used to evaluate the relationships of Sirt6 level to MDA content and SOD activity in patients with adenomyosis.Results Serum Sirt6 level in the normal group was higher than that in the adenomyosis group((7.03±4.59)ng·mL^(-1) vs(2.65±1.52)ng·mL^(-1),P<0.001).The content of MDA in the normal group was lower than that in the adenomyosis group((2.75±2.11)μmol·L^(-1) vs(5.45±2.48)μmol·L^(-1),P<0.001).The activity of SOD in the normal group was higher than that in the adenomyosis group((0.75±0.20)U vs(0.52±0.24)U,P<0.01).The level of Sirt6 was positively correlated with the activity of SOD(r=0.33,P<0.05),but negatively related to the content of MDA(r=-0.56,P<0.05).Conclusion Serum levels of both Sirt6 and oxidative stress increased in patients with adenomyosis.Furthermore,Sirt6 level is correlated with oxidative stress,suggesting that Sirt6 may be involved in the regulation of oxidative damage in adenomyosis.
