摘要
目的研究水通道蛋白1(AQP1)、微小RNA-3195(miR-3195)、PINCH-1在喉癌中的表达及作用机制。方法选取2019年5月至2020年11月收治的81例喉癌患者,对患者癌组织、癌旁组织标本进行免疫组化染色,荧光定量PCR检测AQP1、miR-3195、PINCH-1表达,对比不同病理特征喉癌组织中AQP1、miR-3195、PINCH-1表达情况,分析AQP1、miR-3195、PINCH-1表达在喉癌组织中的相关性。结果喉癌患者癌组织中AQP1、PINCH-1表达高于癌旁组织,miR-3195表达低于癌旁组织(P<0.05)。中低分化程度患者AQP1、PINCH-1表达高于高分化程度患者,miR-3195表达低于高分化程度患者(P<0.05);肿瘤浸润深度为T1~T2期患者AQP1、PINCH-1表达低于T3~T4期患者,miR-3195表达高于T3~T4期患者(P<0.05);TMN分期为Ⅰ~Ⅱ期患者AQP1、PINCH-1表达低于Ⅲ~Ⅳ患者,miR-3195表达高于Ⅲ~Ⅳ患者(P<0.05),淋巴结转移患者AQP1、PINCH-1表达高于淋巴结未转移患者,miR-3195表达低于淋巴结未转移患者(P<0.05)。AQP1、miR-3195、PINCH-1联合检测对喉癌的预测价值优于单一检测。喉癌组织中AQP1、miR-3195呈负相关(r=-0.476,P=0.001);AQP1、PINCH-1呈负相关(r=-0.645,P=0.001);miR-3195、PINCH-1呈正相关(r=0.523,P=0.001)。结论AQP1、miR-3195、PINCH-1在喉癌中的表达出现异常,且AQP1、miR-3195、PINCH-1表达变化与喉癌患者分化程度、肿瘤浸润深度、TNM分期、淋巴结转移情况相关,三者表达之间具有一定相关性,共同参与喉癌的发展过程。
Objective To investigate the expression and action mechanism of aquaporin 1(AQP1),microrNA-3195(Mir-3195)and PINCH-1 in laryngeal carcinoma.Methods A total of 81 patients with laryngeal cancer who were treated in our hospital were enrolled in the study.Immunohistochemical staining was performed for the tumor tissues and para-cancerous tissues of the patients,and the expression levels of AQP1,Mir-3195 and PINCH-1 were detected by fluorescence quantitative PCR.The expressions of AQP1,Mir-3195 and PINCH-1 in laryngeal cancer tissues with different pathological features were observed and compared,and the correlation between the expressions of AQP1,Mir-3195 and pinCH-1 in laryngeal cancer tissues was analyzed.Results The expression levels of AQP1 and PINCH-1 in cancer tissues of patients with laryngeal cancer were significantly higher than those in para-cancerous tissues,however,the expression levels of Mir-3195 were significantly lower than those in para-cancerous tissues(P<0.05).The expression levels of AQP1 and PINCH-1 in patients with medium and low differentiation were significantly higher than those of patients with high differentiation,but,the expression levels of Mir-3195 were significantly lower than those in patients with high differentiation(P<0.05).The expression levels of AQP1 and PINCH-1 in patients with T1-T2 stages were significantly lower than those in patients with T3-T4 stages,however,the mir-3195 levels were significantly higher than those in patients with T3-T4 stages(P<0.05).The expression levels of AQP1 and PINCH-1 in patients with TMN stageⅠ~Ⅱwere significantly lower than those in patients with stageⅢ~Ⅳ,but the expression levels of Mir-3195 were were significantly higher than those in patients with stageⅢ~Ⅳ(P<0.05).Moreover the expression levels of AQP1 and PinCH-1 in patients with lymph node metastasis were were significantly higher than those in patients without lymph node metastasis,however,the expression levels of Mir-3195 were significantly lower than those in patients without lymph node metastasis(P<0.05).In addition the results of combined detection of AQP1,Mir-3195 and PINCH-1 were superior to those by single detection.Furthermore AQP1 and Mir-3195 were negatively correlated in laryngeal carcinoma tissues(P<0.01),and AQP1 and PINCH-1 were negatively correlated(P<0.01),however,Mir-3195 and PINCH-1 were positively correlated(P<0.01).Conclusion The expressions of AQP1,Mir-3195 and PINCH-1 are abnormal in laryngeal cancer,and the expression changes of AQP1,Mir-3195 and PinCH-1 are correlated with the differentiation degree,depth of tumor invasion,TNM stage and lymph node metastasis in patients with laryngeal cancer.The expressions of the three factors are correlated to some extent,which are involved in the development of laryngeal cancer.
