丙泊酚和七氟醚麻醉对老年大鼠腹部术后认知功能的影响 被引量:2

摘要 目的研究丙泊酚和七氟醚两种麻醉药对腹部手术的老年大鼠的认知功能的影响。方法40只大鼠随机分为丙泊酚组和七氟醚组,每组20只。建立丙泊酚和七氟醚麻醉腹部手术大鼠模型,运用Morris水迷宫实验、旷场实验观察大鼠的学习记录能力和情感行为能力。酶联免疫吸附实验、蛋白免疫印迹实验检测大鼠海马炎性因子白介素(IL)-1β、IL-6、肿瘤坏死因子α(TNF-α),氧化应激因子丙二醛(MDA)、超氧化物歧化酶(SOD)和脑源性神经营养因子(BDNF)的蛋白表达。结果丙泊酚组大鼠术后第1、3、5天的逃避潜伏期和游泳距离均显著低于七氟醚组(P<0.05),术后第5天理毛、站立、跨格的次数均显著高于七氟醚组(P<0.05);丙泊酚组大鼠海马中IL-1β、IL-6、TNF-α、MDA的含量均明显低于七氟醚组(P<0.05),SOD的含量和BDNF的蛋白表达均明显高于七氟醚组(P<0.05)。结论丙泊酚麻醉可减轻炎性反应和氧化应激对腹部术后大鼠的认知损害及BDNF的抑制。 Objective To investigate the effects of propofol and sevoflurane anesthesia on the cognitive function of elderly rats after abdominal surgery.Methods Forty rats were randomly divided into propofol group and sevoflurane group,with 20 rats iin eacg group.The animal models of abdominal operation by propofol or sevoflurane anesthesia were established.Morris water maze experiment and open field experiment were used to observe the learning and recording ability and emotional behavior ability of rats.The levels of inflammatory factor Interleukin(IL)-1β,IL-6,tumor necrosis factorα(TNF-α),protein expression of oxidative stress factors malondialdehyde(MDA),superoxide dismutase(SOD)and brain derived neurotrophic factor(BDNF)in hippocampus of rats were detected by enzyme linked immunosorbent assay(ELISA)and Western Blot.Results The escape latency and swimming distance at 1d,3d,5d after operation in propofol group were significantly lower than those in sevoflurane group(P<0.05),however,the times of grooming,standing and crossing grid at 5d after operation were significantly higher than those in sevoflurane group(P<0.05).Moreover the levels of IL-1β,IL-6,TNF-α,MDA in propofol group were significantly lower than those in sevoflurane group,however,the content of SOD and the protein expression levels of BDNF were significantly higher than those in sevoflurane group(P<0.05).Conclusion The propofol anesthesia can relieve the cognitive impairment due to inflammatory reaction and oxidative stress as well as the inhibition of BDNF in rats after abdominal surgery.
