子痫前期相关lncRNA SNHG12对滋养细胞侵袭功能的影响

摘要 目的探讨长链非编码RNA(lncRNA)小核仁RNA宿主基因12(SNHG12)在重度子痫前期患者胎盘组织中的表达,及其对滋养细胞(JEG-3)侵袭功能的影响。方法采用实时荧光定量PCR法(qRT-PCR)检测lncRNA-SNHG12在正常产妇(正常组)及重度子痫前期患者(sPE组)各20例胎盘组织中的表达。JEG-3细胞分为3组:对照组、过表达组和抑制组,分别转染空质粒(pcDNA3.1)、pcDNA3.1-SNHG12和siRNA-SNHG12。采用Transwell试验检测各组细胞侵袭能力;采用qRT-PCR和Western blot检测各组中lncRNA-SNHG12、miR-30a-3p及其靶基因SLUG的表达。结果qRT-PCR结果显示,与正常组织相比,重度子痫前期患者胎盘组织中lncRNA-SNHG12表达量显著降低(P<0.001)。与对照组相比,过表达组lncRNA-SNHG12的水平升高,抑制组水平降低(P<0.001)。Transwell结果显示,与对照组相比,lncRNA-SNHG12过表达组细胞侵袭能力升高,抑制组细胞侵袭能力降低(P<0.001)。与对照组相比,过表达组miR-30a-3p表达降低,抑制组miR-30a-3p表达上升(均P<0.001)。与对照组相比较,过表达组SLUG在mRNA及蛋白水平均升高(P<0.001),抑制组SLUG在mRNA及蛋白水平均降低(P<0.01)。结论lncRNA-SNHG12在子痫前期患者胎盘组织中的表达降低,并引起滋养细胞侵袭能力下降,从而导致子痫前期的发生。 Objective To explore the expression of long non-coding RNA(lncRNA)small nucleolar RNA host gene 12(SNHG12)in placenta of patients with severe preeclampsia and its effect on the invasion function of trophoblast cells(JEG-3).Methods Quantitative real-time PCR(qRT-PCR)was used to detect the expression of lncRNA-SNHG12 in placental tissues of 20 normal parturients(normal group)and 20 severe preeclampsia patients(sPE group).JEG-3 cells were divided into three groups:control group,overexpression group and inhibition group,and transfected with empty plasmid(pcDNA3.1),pcDNA3.1-SNHG12 and siRNA-SNHG12,respectively.Transwell assay was used to detect the invasion ability,and qRT-PCR and Western blot were used to detect the expression of lncRNA-SNHG12,miR-30a-3p and its target gene SLUG in each group.Results The results of qRT-PCR showed that the expression of lncRNA-SNHG12 in placental tissues of severe preeclampsia patients was significantly lower than that of normal parturients(P<0.001).Compared with control group,the level of lncRNA-SNHG12 was increased in overexpression group,and decreased in inhibition group(P<0.001).Transwell assay results showed that compared with control group,the invasion ability of cells was increased in lncRNA-SNHG12 overexpression group,and decreased in inhibition group(P<0.001).Compared with control group,the expression of miR-30a-3p was decreased in overexpression group,and increased in inhibition group(all P<0.001).Compared with control group,the mRNA and protein levels of SLUG were both increased in overexpression group(P<0.001),and decreased in inhibition group(P<0.01).Conclusion LncRNA-SNHG12 is decreased in the placental tissue of patients with preeclampsia,which may lead to preeclampsia by affecting the invasive ability of trophoblast cells.
