摘要
目的 构建一种可以用于肿瘤标志物CA72-4检测的电化学免疫传感器。方法 本实验以硫化钨-还原氧化石墨烯-壳聚糖(WS_(2)-Gr-CS)纳米复合材料修饰电极表面,用纳米金-四氧化三铁-聚多巴胺(Au-Fe_(3)O_(4)-PDA)标记CA72-4二抗,通过银沉积构建了具有多重信号放大的夹心型电化学免疫传感器。结果 在最优条件下,CA72-4在0.0001~5 U·mL^(-1)的浓度范围内,与银溶出电流信号扣除空白值后的增值(ΔI)呈良好的线性关系,检测限为0.00003 U·mL^(-1)。结论 该传感器灵敏度高、抗干扰性强、重现性好,可实现CA72-4的痕量检测,有望用于临床实际血清样品的检测,具有较好的应用前景。
OBJECTIVE To explore an electrochemical immunosensor for the detection of tumor marker CA72-4.METHODS In this experiment, the tungsten sulfide reduced graphene oxide chitosan(WS_(2)-Gr-CS) nanocomposite was used to modify the electrode surface, and the gold ferric oxide polydopamine(Au-Fe_(3)O_(4)-PDA) was used to label CA72-4 secondary antibody.A sandwich electrochemical immunosensor with multiple signal amplification was constructed by silver deposition.RESULTS Under the optimal conditions, the increment of CA72-4 after deducting the blank value from the silver dissolution current signal within the concentration range of 0.0001-5 U·mL^(-1).The detection limit is 0.00003 U·mL^(-1).CONCLUSION The sensor has high sensitivity, strong anti-interference and good reproducibility, and can realize the trace detection of CA72-4.It is expected to be used in the detection of clinical practical serum samples, and has a good application prospect.
