摘要
目的:通过比较抗生素治疗慢性子宫内膜炎前后子宫内膜基因的表达水平,了解慢性子宫内膜炎的治疗对子宫内膜基因谱的影响。方法:19例慢性子宫内膜炎患者采用口服盐酸多西环素片及甲硝唑片治疗,对治疗前后的子宫内膜进行转录组测序,筛选出差异表达基因,并采用GO和KEGG进行生物信息学分析。结果:抗生素治疗慢性子宫内膜炎后相比治疗前有907个基因表达上调,1248个基因表达下调。GO分析结果显示差异基因主要富集于炎症反应调节、急性炎症反应、急性炎症反应调节、细胞—基质黏附、体液免疫反应等生物学过程中,以及细胞外基质部分、细胞外结构部分等细胞成分中;KEGG分析显示差异基因主要富集在IL-17信号通路、PI3K-Akt信号通路、细胞黏附分子、细胞因子—细胞因子受体相互作用、细胞外基质—受体相互作用等通路。结论:抗生素治疗慢性子宫内膜炎前后子宫内膜基因表达存在显著差异,差异表达基因主要富集在炎症反应及免疫反应相关的功能及信号通路,这表明抗生素治疗慢性子宫内膜炎主要通过调节炎症免疫相关基因的表达,从而恢复子宫内膜的功能。
Objective:To understand the effect of chronic endometritis treatment on endometrial gene spectrum by comparing the expression levels of endometrial genes before and after the treatment of chronic endometritis with antibiotic.Methods:19 patients with chronic endometritis were treated with oral doxycycline hydrochloride tablets and metronidazole tablets,then,the endometrium before and after the treatment was subjected to transcriptome sequencing,and differentially expressed genes were screend out,as well as GO and KEGG were used for bioinformatics analysis.Results:After antibiotic treatment for chronic endometritis,there were 907 genes up-regulated and 1248 genes down-regulated compared to before the treatment.The results of GO analysis showed that differential genes were mainly enriched in biological processes such as regulation of inflammatory response,acute inflammatory response,regulation of acute inflammatory response,cell-substrate adhesion,humoral immune response and so on,and cellular components such as extracellular matrix organization,extracellular structure organization and so on;KEGG analysis showed that differential genes were mainly enriched in IL-17 signaling pathway,PI3K-Akt signaling pathway,cell adhesion molecules,cytokine-cytokine receptor interaction,extracellular matrix-receptor interaction and other pathways.Conclusion:There were significant differences in endometrial gene expression between before and after the treatment of chronic endometritis with antibiotic.Differentially expressed genes were mainly enriched in inflammation and immune response-related functions and signal pathways.This indicates that the treatment of chronic endometritis with antibiotic mainly restores the function of the endometrium by regulating the expression of inflammatory and immune-related genes.
