摘要
目的 探讨“益肾温阳,健脾疏肝”背俞穴埋线对围绝经期大鼠肥胖的影响及其机制。方法 48只SD大鼠随机分为空白组、假手术组、模型组和埋线组,每组12只,模型组、埋线组大鼠采用双侧卵巢切除术(OVX)制备围绝经期肥胖大鼠模型,埋线组采用肾俞穴、肝俞穴、脾俞穴埋线治疗。比较各组大鼠体质量变化、Lee′s指数、各部位脂肪重量与重量比,血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C),棕色脂肪组织(BAT)中环磷酸腺苷(cAMP)含量及解偶联蛋白1(uncoupling protein, UCP1)、过氧化物酶增殖活化受体γ协同刺激因子1(PGC1)、氧化物酶增殖激活受体γ(PPARγ)mRNA与蛋白表达。结果 与假手术组相比,模型组大鼠体质量、Lee′s指数、各部位脂肪重量与白色脂肪重量比明显升高,血清TC、TG、LDL-C含量升高、HDL-C含量下降,BAT中cAMP含量及UCP1、PGC1、PPARγmRNA与蛋白表达下调,差异具有统计学意义(P<0.05,P<0.01)。经穴位埋线治疗后,大鼠体质量、Lee′s指数、各部位脂肪重量与白色脂肪重量比降低,血清TG、LDL-C含量降低、HDL-C含量升高,BAT中cAMP含量、UCP1、PGC1、PPARγmRNA与蛋白表达上调,差异具有统计学意义(P<0.05,P<0.01)。结论 “益肾温阳,健脾疏肝”穴位埋线能够防治围绝经期大鼠肥胖,其机制可能是通过增加BAT内cAMP含量,上调UCP1、PGC1、PPARγmRNA与蛋白表达,加强脂肪组织能量代谢实现的。
Objective To investigate the effect and the mechanism of “tonifying kidney and warming Yang, strengthening spleen and soothing liver” catgut embedding at back Shu acupoints on obesity in perimenopausal rats.Methods Forty-eight SD rats were randomly divided into blank group, sham operation group, model group and catgut embedding group, 12 rats in each group.Model group and catgut embedding group were treated with bilateral ovariectomy(OVX)to prepare the perimenopausal obesity model.The catgut embedding group was treated with Shenshu(BL23),Ganshu(BL18)and Pishu(BL20).The body weight changes, Lee′s index, fat weight to weight ratio of each part, serum levels of total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)were compared in each group, cAMP content and uncoupling protein(UCP1),peroxisome Proliferator-activated receptors γ(PPARγ),peroxisome proliferator activated receptor 1(PGC1)mRNA and protein in brown adipose tissue(BAT)were compared.Results Compared with those of the sham operation group, the body weight, Lee′s index, fat weight ratio of each part and white fat weight ratio of model group increased significantly, the serum levels of TC,TG and LDL-C increased and the lerel of HDL-C decreased, cAMP content and UCP1,PGC1,PPARγ mRNA and protein in BAT were down-regulated, which were significantly different(P<0.05,P<0.01).After catgut embedding therapy, the body weight, Lee′s index, fat weight ratio of each part and white fat weight ratio of rats decreased, serum levels of TG and LDL-C decreased, HDL-C level increased, cAMP content, UCP1,PGC1,PPARγ mRNA and protein in BAT were up-regulated and the difference was significant(P<0.05,P<0.01).Conclusion The mechanism of catgut embedding can prevent and cure obesity in perimenopausal rats.The mechanism may be to increase cAMP content in BAT and increase UCP1,PGC1,PPARγ mRNA and protein expressions and enhance the energy metabolism of fat tissue.
