复方蜥蜴散凝胶对溃疡性结肠炎模型大鼠Claudin-1和Zonulin蛋白表达的影响 被引量:2

摘要 目的 研究复方蜥蜴散凝胶对溃疡性结肠炎(UC)大鼠结肠组织Claudin-1和Zonulin蛋白的表达影响,探究其治疗UC的机制。方法 选取SD雄性大鼠120只,随机分为空白对照组和模型组,模型组以2,4,6-三硝基苯磺酸(TNBS)造模,造模成功后,随机分为复方蜥蜴散凝胶低、中、高剂量组,柳氮磺吡啶组,模型对照组。给予复方蜥蜴散凝胶不同浓度、柳氮磺吡啶、生理盐水,治疗14 d,观察大鼠一般情况;干预后麻醉处死,取病变结肠组织,观察结肠形态学变化,Western blot法和免疫组化法(IHC)检测大鼠结肠组织中Claudin-1、Zonulin蛋白的表达,统计分析实验结果。结果 模型对照组与空白对照组比较:大鼠结肠上皮细胞受损严重、腺体破坏、出现溃疡等病变,并伴有大量炎性细胞浸润。复方蜥蜴散凝胶高、中、低剂量组及柳氮磺吡啶组较模型组:大鼠肠上皮细胞损伤程度减轻,溃疡灶数减少,溃疡面积缩小,炎性细胞浸润减少。模型对照组与空白对照组比较,大鼠结肠组织中Claudin-1蛋白表达水平降低(P<0.05),Zonulin蛋白表达水平升高(P<0.05);柳氮磺吡啶组及各治疗组与模型对照组比较,Claudin-1蛋白表达水平明显升高(P<0.05),Zonulin蛋白表达水平明显降低(P<0.05)。结论 复方蜥蜴散凝胶可以通过影响蛋白表达水平,修复损伤结肠黏膜,发挥治疗UC的作用。 Objective To study the effect of Compound Lizard Powder Gel on the expressions of Claudin-1 and Zonulin proteins in colonic tissue of ulcerative colitis(UC)rats and explore the mechanism of UC treatment.Methods A total of 120 male SD rats were randomly divided into blank control group and model group.The model group was modeled with 2,4,6-three nitrobenzene sulfonic acid(TNBS).After successful modeling, the rats were randomly divided into low, medium and high dose groups of Compound Lizard Powder Gel, sulfasalazine group and model control group.The rats were treated with different concentrations of Compound Lizard Powder Gel, sulfasalazine and normal saline for 14 days, and the general condition of rats was observed.After the intervention, the rats were killed under anesthesia, and the diseased colon tissues were taken out to observe the morphological changes of colon.The expressions of Claudin-1 and Zonulin proteins in rat colon tissues were detected by Western blot and immunohistochemistry(IHC),and the experimental results were statistically analyzed.Results Compared with the blank control group, the model control group had severe damage to colonic epithelial cells, gland destruction, ulcer and the infiltration of a large number of inflammatory cells.Compared with the model group, the damage rate of intestinal epithelial cells was reduced, the number of ulcerated foci decreased, the area of ulcer reduced, and the infiltration of inflammatory cells decreased.Western blot and IHC were used to analyze the expressions of Claudin-1 and Zonulin proteins in rat colon.Compared with those of the blank control group, the expressions of Claudin-1 protein in rat colon decreased(P<0.05),and the expressions of Zonulin protein increased(P<0.05).Compared with those of the model control group, the expressions of Claudin-1 protein increased significantly(P<0.05) and the expressions of Zonulin protein decreased significantly(P<0.05).Conclusion Compound Lizard Powder Gel can affect the protein expressions and repair the damaged colonic mucosa, so as to play a role in the treatment of UC.
