丹瓜方对糖尿病大鼠胰腺形态及GLUT4、LEP表达的影响 被引量:1

Effects of Dangua Decoction(丹瓜方)on Pancreatic Morphology and Expression of GLUT4 and LEP in Diabetic Rats

摘要 目的探讨丹瓜方对糖尿病大鼠胰腺形态及葡萄糖转运蛋白4(glucose transporter 4,GLUT4)、瘦素(leptin,LEP)表达的影响。方法SD大鼠按前期研究方案造模成糖尿病大鼠,随机分为正常组、模型组、二甲双胍组、常规剂量组、高剂量组。干预16周后分离获取胰腺组织并进行HE染色,检测糖尿病大鼠肌肉和脂肪组织GLUT4、LEP的表达。结果HE染色形态:正常组胰岛细胞外观呈饱满球形,细胞排列匀齐,胞质内含有大量胰岛素分泌颗粒,胰岛细胞占胰岛大部分范围;胰岛数量较多,面积较大,均衡分布并和腺泡分界清楚。与正常组比较,模型组胰岛细胞形态萎缩,排列混乱;胞质内胰岛素分泌颗粒显著减少;胰岛数量较少,面积较小,不均匀分布。与模型组比较,常规剂量组、高剂量组及二甲双胍组胰岛结构较规整,胰岛面积较大,胰岛细胞萎缩程度较轻,胰岛素分泌颗粒较多。与正常组比较,模型组和二甲双胍组大鼠肌肉GLUT4蛋白的表达量显著降低(P<0.01),常规剂量组大鼠肌肉GLUT4蛋白表达显著增加(P<0.01);模型组大鼠脂肪LEP的mRNA表达量显著降低(P<0.01)。和模型组相比,常规剂量组、高剂量组的大鼠肌肉GLUT4蛋白表达显著增加(P<0.01),二甲双胍组、常规剂量组、高剂量组大鼠脂肪LEP的mRNA表达量增加(P<0.05或P<0.01);与二甲双胍组比较,常规剂量组及高剂量组大鼠肌肉GLUT4蛋白表达显著增加(P<0.01),常规剂量组大鼠脂肪LEP的mRNA表达量显著增加(P<0.05)。结论丹瓜方可改善胰岛细胞的形态,丹瓜方可以通过调节GLUT4蛋白及LEP的mRNA表达来影响胰岛素功能达到控制血糖的目的。 Objective To investigate the effect of Dangua Decoction(丹瓜方)on pancreatic morphology and expression of glucose transporter 4 and leptin in diabetic rats.Methods SD rats were modeled as diabetic rats according to the previous study protocol and then divided into normal group,model group,metformin group,conventional-dose Dangua Decoction group,and high-dose Dangua Decoction group randomly.After 16 weeks of intervention,pancreatic HE staining was performed to detect the expression of GLUT4 and LEP in muscle and adipose tissue of diabetic rats.Results HE staining and morphological observation:In the normal group,the islet cells were full spherical and arranged neatly,a large number of insulin-secreting granules were observed in the cytoplasm,accounting for most of the range of islets.The number of islets and the area were both large,the distribution was uniform,and the acini were clearly demarcated.Compared with the normal group,the islet cells were atrophic and disorganized,the insulin secretory granules in the cytoplasm were significantly cut down.The number of islets in the model group and the area were both reduced,the distribution was sparse and uneven.Comparing to the model group,the islet structure was more regular,the islet area was larger,the degree of islet cell atrophy was milder,and there were more insulin-secreting granules in the metformin,conventional-dose and high-dose groups.Compared with the normal group,the expression of GLUT4 protein in the muscle of rats in the model group and metformin group were decreased(P<0.01),the expression in the conventional-dose group was increased(P<0.01),and the mRNA expression of LEP in the fat of rats in the model group was significantly decreased(P<0.01).Comparing with the model group,the expression of GLUT4 protein in the muscle of rats in the conventional-dose and high-dose groups were increased(P<0.01),and the mRNA expression of LEP in the fat of rats in the metformin,conventional-dose and high-dose groups were increased(P<0.05 or P<0.01).Compared with the metformin group,the expression of GLUT4 protein in the muscle of rats in the conventionaldose and high-dose groups increased(P<0.01),and the mRNA expression of LEP in the fat of rats in the conventional-dose group was increased(P<0.05).Conclusion Dangua Decoction can improve the morphology of islet cells,and also can affect insulin function by regulating the expression of GLUT4 protein and the mRNA expression of LEP to achieve the purpose of controlling blood glucose.
