摘要
目的研究百部多糖对黄芩苷及其代谢产物黄芩素6-O-β-D-葡萄糖醛酸苷(B6G)药代动力学影响。方法大鼠随机分为两组,口服灌胃给药后眼眶后静脉丛取血。利用超高效液相色谱联用四级杆串联飞行时间质谱仪(UPLC-Q-TOF-MS/MS)技术鉴定血浆中黄芩苷与B6G。采用超高效液相色谱联用三重四级杆质谱仪(UPLC-TQ-MS/MS)结合多反映监测(MRM)模式测定血浆中黄芩苷和B6G的浓度,运用DAS 2.0软件处理药代动力学参数。结果经色谱图与质谱图可知,黄芩苷与B6G互为同分异构体。药代动力学结果可知,与黄芩苷组相比较,黄芩苷+百部多糖组的AUC0-t显著增加,提高了1.32倍,而B6G的药代动力学参数没有显著性变化。结论百部多糖能增加体内黄芩苷的血药浓度,促进黄芩苷的口服生物利用度。
Objective To study the effects of stemona polysaccharides on the pharmacokinetics of baicalin and its metabolite baicalein 6-O-β-D-glucuronide(B6G).Methods The rats were randomly divided into two groups.After oral gavage,blood was taken from the retro-orbital venous plexus.UPLC-Q-TOF-MS/MS technology was used to identify baicalin and B6G in plasma.UPLCTQ-MS/MS combined with multiple reaction monitoring(MRM)mode was used to determine the concentration of baicalin and B6G in plasma,using DAS2.0 software deal with pharmacokinetic parameters.Results According to chromatogram and mass spectrum,it can be seen that baicalin and B6G are isomers of each other.The pharmacokinetic results showed the AUC0-t of baicalin+stemona polysaccharides increased significantly by 1.32 times,compared with the baicalin group,while the pharmacokinetic parameters of B6G didn’t change significantly.Conclusion Stemone polysaccharides can increase the plasma concentration of baicalin in the body and promote the oral bioavailability of baicalin.
