电针对功能性消化不良模型大鼠环氧合酶2介导炎症反应机制研究 被引量:3

Effect of Electroacupuncture on Cyclooxygenase-2 Mediated Inflammatory Response in Functional Dyspepsia Model Rats

摘要 目的观察电针足三里配中脘穴是否通过抑制十二指肠中环氧合酶2(COX-2)、P物质(SP)、降钙素基因相关肽(CGRP)、白细胞介素6(IL-6)的表达,从而对功能性消化不良(FD)炎症反应起到干预作用。方法选取48只10日龄幼年SD大鼠,雌雄各半,随机分为正常组、模型组、电针组、非经非穴组,每组12只,采用碘乙酰胺灌胃法制备FD大鼠模型。电针组电针足三里配中脘穴位,1次/d,连续14 d,非经非穴组进行电针1次/d,连续14 d。用免疫组化法和定量PCR法检测十二指肠中COX-2的蛋白及mRNA表达,用ELISA方法检测十二指肠中SP、CGRP、IL-6含量的表达。结果与正常组比较,模型组大鼠十二指肠中COX-2蛋白和mRNA表达均显著升高(P<0.01),且十二指肠中SP、CGRP、IL-6含量表达显著升高(P<0.01);非经非穴组大鼠的COX-2、SP、IL-6与正常组比较表达显著升高(P<0.01),CGRP的表达有所升高(P<0.05);与模型组比较,通过电针足三里、中脘干预后的大鼠十二指肠中COX-2蛋白和mRNA表达降低(P<0.05),且十二指肠中SP、CGRP、IL-6含量表达水平降低(P<0.05);电针组与非经非穴组比较有差异,电针组大鼠十二指肠中COX-2的mRNA含量表达与SP、CGRP、IL-6各含量表达较非经非穴组有所降低(P<0.05)。结论电针足三里配中脘穴可能通过降低COX-2基因的表达而抑制炎症反应的发生,从而起到对FD干预作用。 Objective To observe whether electroacupuncture Zusanli(ST36)and Zhongwan(CV12)could inhibit the expression of cyclooxygenase-2(COX-2),substance P(SP),calcitonin gene-related peptide(CGRP)and interleukin-6(IL-6)in duodenal tissue.Thus it can intervene the inflammatory response of functional dspepsia(FD).Methods Choose forty-eight 10-day-old young SD rats were randomly divided into normal group,model group,electroacupuncture group,non-acupuncture and nonacupuncture group,with 12 rats in each group.The FD rat model was prepared by intragastric administration of iodoacetamide.The electroacupuncture group received electroacupuncture at Zusanli(ST36)and Zhongwan(CV12)acupoints,once a day,for consecutive 14 days,and the non-acupuncture and non-point groups received electroacupuncture once a day,for consecutive 14 days.The protein and mRNA expression of COX-2 in duodenum were detected by immunohistochemistry and quantitative PCR,and the content of SP,CGRP and IL-6 in duodenum was detected by ELISA.Results Compared with normal group,the expression of COX-2 protein and mRNA in duodenum of model group was significantly increased(P<0.01),and the content of SP,CGRP and IL-6 in duodenum of model group was significantly increased(P<0.01).The expression of COX-2,SP and IL-6 in non-acupoint group was significantly higher than that in normal group(P<0.01),and the expression of CGRP was increased(P<0.05).Compared with model group,the expression of COX-2 protein and mRNA in duodenum of Zusanli Zhongwan intervention rats decreased(P<0.05),and the expression levels of SP,CGRP and IL-6 in duodenum decreased(P<0.05).The expression of COX-2 mRNA and the content of SP,CGRP,IL-6 in duodenum of electroacupuncture group were lower than those in non-acupuncture and non-acupuncture group(P<0.05).Conclusion Electroacupuncture Zusanli(ST36)and Zhongwan(CV12)may inhibit the occurrence of inflammatory response by reducing the expression of COX-2 gene,thus playing an intervention role in FD.
