摘要
为探究不同萃取相黄果茄果实乙醇提取物的生物活性,本试验采用自由基清除法、D-半乳糖衰老模型、热板法和小鼠耳廓肿胀模型分别检测了黄果茄果实乙醇提取物石油醚萃取相(PFSX)、二氯甲烷萃取相(DFSX)、乙酸乙酯萃取相(EFSX)、正丁醇萃取相(NFSX)和水萃取相(WFSX)五个部位的抗氧化活性、镇痛活性和抗炎活性。结果显示,不同极性部位对DPPH·的清除能力为EFSX>NFSX>DFSX>WFSX>PFSX,各相均能提高D-半乳糖衰老模型小鼠血清的总抗氧化能力(T-AOC)和超氧化物歧化酶(SOD)活力;给药30 min后,DFSX、NFSX和WFSX显著提高小鼠的痛阈值(P<0.05或P<0.01),给药60 min后,PFSX、DFSX、NFSX和WFSX显著提高小鼠的痛阈值(P<0.05或P<0.01),并且对热板所致疼痛有不同程度的抑制作用(P<0.05或P<0.01);PFSX、DFSX和EFSX能极显著抑制二甲苯所致小鼠耳廓肿胀(P<0.01),WFSX能显著抑制二甲苯所致小鼠耳廓肿胀(P<0.05);WFSX、DFSX和EFSX能极显著降低组织中COX-2含量(P<0.01),DFSX和EFSX极显著降低组织中TNF-α和IL-6含量(P<0.01)。结果表明,不同萃取相黄果茄果实乙醇提取物均具有抗氧化、镇痛和抗炎活性,其中EFSX的抗氧化活性最好,DFSX和EFSX具有良好的抗炎活性。本试验结果为黄果茄果实的药理研究与产品研发提供了一定的理论支持。
In order to determine the biological activity of ethanol extracts from Solanum xanthocarpum fruit, petroleum ether fraction(PFSX), dichloromethane fraction(DFSX), ethyl acetate fraction(EFSX), N-butanol fraction(NFSX) and water fraction(WFSX) of Solanum xanthocarpum ethanol extracts were examined for antioxidant, analgesic and anti-inflammatory activities by free radical scavenging method, D-galactose aging model, hot plate method and mouse auricle swelling model. The results showed the DPPH· scavenging capacity of different phases in the free radical scavenging experiment was in the order of EFSX>NFSX>DFSX>WFSX>PFSX, each extract phase increased the activity of total antioxidant capacity(T-AOC)、superoxide dismutase(SOD) in D-galactose ageing model mouse serum. After 30 min of administration, DFSX, NFSX and WFSX significantly prolonged the pain threshold of mice(P<0.05 or P<0.01). After 60 min of administration, PFSX, DFSX, NFSX and WFSX clearly prolonged the pain threshold of mice(P<0.05 or P<0.01) and inhibited the pain caused by hot plate in different degrees(P<0.05 or P<0.01);PFSX, DFSX and EFSX most significantly inhibited xylene-induced ear swelling(P<0.01), while WFSX significantly inhibited xylene-induced ear swelling(P<0.05). WFSX, DFSX and EFSX significantly decreased the contents of COX-2 in inflammatory tissue(P<0.01), while DFSX and EFSX significantly decreased the contents of TNF-α and IL-6 in inflammatory tissue(P<0.01). Thus, the different phases of ethanol extracts of Solanum xanthocarpum fruit display antioxidant, analgesic and anti-inflammatory activities, the antioxidant activity of EFSX from ethanol extract appears to the best, and DFSX and EFSX have good anti-inflammatory activity. This study lays a sound pharmacological basis for further development and application of ethanol extracts of Solanum xanthocarpum fruit.
