响应面法优化超声协同胃蛋白酶提取鲣鱼皮胶原蛋白的工艺研究 被引量:1

摘要 以鲣鱼皮为原料,利用响应面法对超声协同胃蛋白酶提取鲣鱼皮胶原蛋白的工艺进行优化。在单因素试验的基础上,以鲣鱼皮胶原蛋白提取率为响应值,选择超声时间、酶解温度、酶解时间和酶用量进行Box-Behnken试验设计,对影响胶原蛋白提取率的各工艺条件及其交互作用的影响进行研究,得到了二次多项式回归方程模型。结果表明:鲣鱼皮胶原蛋白的最佳提取工艺条件为超声时间146 min、酶解温度36℃、酶解时间249 min、酶用量183 U/g,重复3次实验作平行验证,得到鲣鱼皮胶原蛋白提取率的平均值为38.11%,与预测值的相对误差为0.37%,说明该回归模型具有一定的可靠性,准确度好,该研究为鲣鱼皮开发成胶原蛋白及相关产品提供参考。 Using skipjack tuna skin as raw material,the technology of ultrasonic synergistic pepsin extraction of skipjack tuna skin collagen was optimized by response surface methodology.On the basis of single factor experiment,taking the extraction yield of skipjack tuna skin collagen as the response value,the Box Behnken experiment was designed by selecting ultrasonic time,enzymolysis temperature,enzymolysis time and enzyme dosage.The effects of various technological conditions affecting the extraction yield of collagen and their interactions were studied,and a quadratic polynomial regression equation model was obtained.The results showed that the optimal extraction conditions of skipjack tuna skin collagen were ultrasonic time 146 min,enzymolysis temperature 36℃,enzymolysis time 249 min,and enzyme dosage 183 U/g.Under these conditions,the extraction yield of skipjack tuna skin collagen was 38.11%,and the relative error with the predicted extraction yield was 0.37%,indicating that the regression model had certain reliability and good accuracy.This study provided a reference for the development of skipjack tuna skin into collagen and related products.
