一株猪圆环病毒3型的鉴定和分子特征分析

摘要 为了解猪圆环病毒(PCV)3型的基因组特征和遗传变异情况。本试验利用PCR方法从病死猪脾脏中鉴定出一株PCV3,命名为SC-YB2108。经过PCR方法扩增全序列,对测序结果拼接并进行系统进化树和遗传重组分析。结果显示:SC-YB2108序列长度2 000 bp;遗传进化树分析发现SC-YB2108全基因组与中国株MK142771,MG96866,美国株MK496279,KX458235,韩国株MH231553处于同一独立分支,属于PCV3f。序列分析发现SC-YB2108与广西株MK095620同源性为99.2%,与湖南株MG372488的同源性为91.6%,与四川株MZ449244和OK334614的同源性分别为98.4%和98.7%。本研究有助于深入了解四川省PCV3流行毒株的分子特性,为后续PCV3的诊断试剂和疫苗开发提供依据。 To understand the genomic characteristics and genetic variation of circovirus(PCV)type 3.In this experiment,a PCV3 strain named SC-YB2108 was identified from the spleen of dead pigs by PCR.The whole sequence was amplified by PCR method,and the sequencing results were spliced for phylogenetic tree and genetic recombination analysis.The results showed that the length of SC-YB2108 sequence was 2 000 bp;Genetic evolution tree analysis showed that the whole genome of SC-YB2108was in the same independent branch as that of Chinese strain MK142771,MG96866,American strain MK496279,KX458235and Korean strain MH231553,belonging to PCV3f.Sequence analysis showed that SC-YB2108 had 91.6% homology with Guangxi strain MK095620(99.2%),Hunan strain MG372488,and 98.4% and 98.7% homology with Sichuan strain MZ449244and OK334614,respectively.This study will help to further understand the molecular characteristics of PCV3 epidemic strains in Sichuan Province,and provide a basis for the subsequent development of diagnostic reagents and vaccines for PCV3.
