摘要
目的:探讨槲皮素对顺铂所致大鼠卵巢损伤的保护作用。方法:选用雌性SD大鼠32只,随机分为4组,每组8只:正常对照组、顺铂损伤模型组、槲皮素低浓度保护组及槲皮素高浓度保护组。顺铂损伤模型组与槲皮素保护组均腹腔注射顺铂2.0 mg/(kg·d),连续1周,槲皮素低浓度保护组50 mg/(kg·d)、槲皮素高浓度保护组100 mg/(kg·d)槲皮素灌胃处理2周。正常对照组大鼠相同剂量生理盐水处理。结果:槲皮素能保护顺铂引起的大鼠体重下降,与正常对照组效应相近。顺铂显著降低大鼠血清中E2、P4的含量(P <0.01),槲皮素保护组雌孕激素水平较模型组升高(P <0.05)。顺铂损伤模型组卵巢颗粒细胞数量少,凋亡细胞明显多于活细胞,而槲皮素保护组凋亡细胞数量明显少于顺铂损伤模型组(P <0.05)。结论:顺铂对大鼠卵巢有明显损伤作用,槲皮素在一定程度上能保护顺铂引起的卵巢损伤。
Objective:To investigate the protective effect of quercetin(Q)on cisplatin-induced ovarian injury in rats.Methods:32 female SD rats were selected and randomLy divided into 4 groups of 8 rats each:Normal control group、ovarian damage model group、Q low and high concentration protection group.Both the ovarian injury model group and the Q-protected group were injected intraperitoneally with cisplatin 2.0 mg/(kg·d)for one week.Q low concentration protection group 50 mg/(kg·d),Q high protection group 100 mg/(kg·d)Q gavage treatment for two weeks.Normal control rats were treated with the same dose of saline.Results:Q protected against cisplatin-induced weight loss in rats,with similar effects to normal controls.Cisplatin reduced the levels of E2 and P4 in the serum of rats(P < 0.01),and the estrogen and progesterone levels were increased in the Q-protected group compared with the model group(P < 0.05).The number of ovarian granulosa cells in the cisplatin-injured model group was low and the number of apoptotic cells was more than that of live cells,while apoptotic cells in the Q-protected group was less than that of the cisplatin-injured model group(P < 0.05).Conclusion:Cisplatin could damage on rat ovaries,and Q could protect cisplatin-induced ovarian failure.
