不同配比生姜-黄芩药对对胃寒证模型大鼠能量代谢酶、胃肠动力及脑肠肽的影响 被引量:1

Effects of Shengjiang(Rhizoma Zingiberis Recens)-Huangqin(Radix Scutellariae) Herbal Pair at Different Composition Ratios on Energy Metabolism Enzymes, Gastrointestinal Motility and Brain-gut Peptides in Rats with Stomach Cold Syndrome

摘要 目的 观察生姜、黄芩不同比例配伍对胃寒证的影响,并探讨可能的作用机制。方法 SD大鼠随机分为空白组12只和造模组60只。采用“0℃纯净水灌胃+冷水泡浴+氢氧化钠刺激”的方法建立大鼠胃寒证模型,造模18天。将造模成功的50只大鼠随机分为模型组、生姜组、生姜黄芩1∶1组、生姜黄芩5∶3组、生姜黄芩2∶1组,每组10只。空白组与模型组灌胃生理盐水10 ml/kg,生姜组灌胃生姜水煎液3.6 g/kg,生姜黄芩1∶1组、5∶3组、2∶1组分别灌胃相应水煎液7.2、9.6、10.8 g/kg,各组均每天灌胃1次,连续7天。观察大鼠一般状况。检测各组大鼠肝组织能量代谢酶葡萄糖激酶(GCK)、丙酮酸脱氢酶(PDH)、异柠檬酸脱氢酶(ICD)、Na~+-K~+-三磷酸腺苷酶(Na~+-K~+-ATP)、脂肪甘油三酯脂肪酶(ATGL)含量;观察各组大鼠胃组织病理形态,计算胃排空率和小肠推进率,测定胃组织脑肠肽胃动素(MTL)、5-羟色胺(5-HT)、胃泌素(GAS)、生长抑素(SS)、血管活性肠肽(VIP)含量。结果 与空白组比较,模型组大鼠胃黏膜结构不完整,黏膜组织坏死脱落,腺体排列紊乱,可见大量炎性细胞浸润;与模型组比较,生姜黄芩1∶1组局部黏膜坏死脱落,炎细胞浸润;其余各给药组胃黏膜上皮结构完整,腺体排列整齐,未见明显炎细胞浸润。与空白组比较,模型组大鼠肝组织GCK、PDH、ICD及Na~+-K~+-ATP的含量,胃排空率、小肠推进率显著降低,胃组织5-HT、MTL、GAS含量显著降低,SS、VIP含量显著升高(P<0.01)。与模型组比较,生姜组及生姜黄芩5∶3、2∶1组肝组织GCK、PDH、ICD及Na~+-K~+-ATP的含量,胃排空率,小肠推进率及胃组织5-HT、MTL、GAS含量显著升高,SS、VIP含量显著降低;生姜黄芩1∶1组肝组织PDH、ICD的含量显著升高,胃组织SS、VIP含量显著降低(P<0.05或P<0.01)。与生姜组比较,生姜黄芩5∶3组肝组织GCK、PDH、Na~+-K~+-ATP的含量,胃排空率、小肠推进率,胃组织5-HT、MTL、GAS含量显著升高,VIP含量显著降低;生姜黄芩2∶1组肝组织GCK、PDH、ICD、Na~+-K~+-ATP的含量,小肠推进率显著升高(P<0.05或P<0.01)。与生姜黄芩5∶3组比较,生姜黄芩2∶1组肝组织GCK、PDH、ICD,胃组织VIP显著升高,胃排空率,胃组织5-HT、MTL、GAS含量显著降低(P<0.05或P<0.01)。结论 不同配比生姜、黄芩水煎液均能通过调节能量代谢酶活性以及脑肠肽含量等机制,不同程度地治疗胃寒证,以生姜、黄芩5∶3配伍效果更佳。 ObjectiveTo observe the effects of Shengjiang(Rhizoma Zingiberis Recens)-Huangqin(Radix Scutellariae)herbal pair at different composition ratios in the treatment of stomach cold syndrome,and to explore itspotential mechanism.MethodsSD rats were randomly divided into blank group(n=12)and model group(n=60).The rat model of stomach cold syndrome was established by the method of gavage with pure water at 0 ℃ plus cold wa-ter bath and sodium hydroxide stimulation for 18 days. Fifty successfully modeled rats were randomly divided intomodel group,Shengjiang group,Shengjiang-Huangqin(S-H)1∶1,5∶3,and 2∶1 group,with 10 rats in eachgroup. The blank group and the model group were gavaged with 10 m L/kg of normal saline,while Shengjiang groupwas gavaged with 3. 6 g/kg of Shengjiang decoction,and S-H 1∶1,5∶3 and 2∶1 groups were gavaged with 7. 2,9. 6 and 10. 8 g/kg of corresponding S-H decoction,respectively. All groups were gavaged once a day for 7 consecu-tive days. The general condition of rats was observed. The contents of energy metabolizing enzymes glucokinase(GCK),pyruvate dehydrogenase(PDH),isocitrate dehydrogenase(ICD),Na-K-ATPase(Na-K-ATP)and adi-pose-triglyceride lipase(ATGL)in liver tissue of rats in each group were detected. The pathological morphology ofgastric tissue was observed,and the gastric emptying rate and small intestinal propulsion were calculated. The con-tents of motilin(MTL),5-hydroxytryptamine(5-HT),gastrin(GAS),somatostatin(SS)and vasoactive intestinalpeptide(VIP)in gastric tissue were determined.ResultsCompared to those in the blank group,the gastric mucosaof the rats in the model group was incomplete,and the mucosal tissue was necrotic and exfoliated,with disorderly ar-ranged glands and a large number of infiltrated inflammatory cells. Compared to those in the model group,local muco-sal necrosis and shedding,as well as inflammatory cell infiltration were seen in the S-H 1∶1 group. The gastric muco-sa epithelial structure was complete,and the glands were neatly arranged in other medicinal groups,with no obviousinflammatory cell infiltration. Compared to those in the blank group,the contents of GCK,PDH,ICD and Na-K-ATP in liver tissue,gastric emptying rate and small intestinal propulsiion,as well as the contents of 5-HT,MTL andGAS were significantly reduced in the model group,while the contents of SS and VIP significantly increased(P< 0. 01). Compared to those in the model group,the contents of GCK,PDH,ICD and Na-K-ATP in liver tissue,gas-tric emptying rate and small intestinal propulsion,as well as the contents of 5-HT,MTL and GAS in stomach tissuesignificantly increased,while the contents of SS and VIP significantly decreased in Shengjiang group,S-H 5∶3 and 2∶1 groups;the contents of PDH and ICD in liver tissue significantly increased,while the contents of SS and VIP instomach tissue significantly decreased in the S-H 1∶ 1 group(P<0. 05 or P<0. 01). Compared to those in theShengjiang group,the contents of GCK,PDH,ICD,and Na-K-ATP in liver tissue,gastric emptying rate,small in-testinal propulsion,and the contents of 5-HT,MTL and GAS in stomach tissue significantly increased,while the VIPcontent significantly decreased in S-H 5∶3 group;the contents of GCK,PDH,and Na-K-ATP in liver tissue,andsmall intestinal propulsion significantly increased in S-H 2∶1 group(P<0. 01 or P<0. 01). S-H 2∶1 group hadhigher GCK,PDH,ICD contents in liver tissue and VIP content in stomach tissue,as well as lower gastric emptyingrate and contents of 5-HT,MTL and GAS in stomach tissue than S-H 5∶3 group(P<0. 05 or P<0. 01).ConclusionS-H decoction at different composition ratios all have effects on stomach cold syndrome by regulating the activi-ty of energy metabolism enzyme and the content of brain-gut peptide. The effect is varied,and S-H at the ratio of 5∶3may be the best.
