摘要
褪黑素作为一种植物激素,能够影响种子萌发,起到抗氧化剂的作用。本实验以荻为实验材料,在培养皿中进行培养,并设置不同褪黑素处理浓度(0,1,10,50,100,500μM),通过萌发指标、生长指标和一些生理指标的测定,研究无胁迫条件下褪黑素对荻种子萌发及幼苗生长和生理的影响。结果表明:随着褪黑素的加入,荻种子的萌发状态变好,其中,10μM和50μM的褪黑素对荻种子萌发和幼苗生长的促进效果较好;50μM褪黑素对荻种子的萌发指标、荻幼苗的苗高和过氧化物酶活性的促进效果最好,且50μM褪黑素处理下的荻幼苗的电导率最低,说明细胞被破坏的程度最小;10μM褪黑素对荻种子萌发及幼苗生长的综合促进效果最好,它对荻种子的根长、鲜重、淀粉酶活性、脂肪酶活性、叶绿素含量、类胡萝卜素含量、超氧化物歧化酶活性和过氧化氢酶活性的促进效果最显著。该研究可为后期将褪黑素对种子萌发的促进作用应用于农业生产提供理论基础。
As a plant hormone,melatonin can affect seed germination and act as an antioxidant.In this experi-ment,Miscanthus sacchariflorus was used as the experimental material,culturing in a petri dish,and different con-centrations of melatonin(1,10,50,100,500μM)were set to study the effect of melatonin on seed germination,seedling growth and physiology of Miscanthus sacchariflorus by the determinations of germination indicators,growth indicators and some physiological indicators.It was found that with the add of melatonin concentration,the germination status of Miscanthus sacchariflorus seeds became better,and the melatonin concentrations of 10μM and 50μM had good promotion on seed germination and seedling growth.Under the treatment of melatonin concentration of 50μM,the germination indicators,seedling height and peroxidase activity of Miscanthus sacchar-iflorus seedlings had the best promotion effect,and the conductivity of Miscanthus sacchariflorus seedlings was the lowest,indicating that the degree of cell damage was the least.The melatonin concentration of 10μM had the best promotion on seed germination and seedling growth.This treatment group had the most significant promoting effects on the root length,fresh weight,amylase activity and lipase activity,the contents of chlorophyll and carot-enoid,superoxide dismutase activity and catalase activity of Miscanthus sacchariflorus.This study can provide a theoretical basis for the later application of melatonin to promote seed germination in agricultural production.
