摘要
目的比较不同剂量木犀草素在大鼠体内的药动学。方法大鼠随机分为6组,分别灌胃给予10、20、40、80、120 mg/kg木犀草素及尾静脉注射给予木犀草素注射液(4 mg/kg),于不同时间点采血,HPLC法测定木犀草素血药浓度,计算主要药动学参数。结果在10~80 mg/kg剂量范围内,AUC_(0~∞)、C_(max)与剂量的相关系数分别为0.9966、0.9694。当剂量为120 mg/kg时,C_(max)不再增加,AUC_(0~∞)与剂量的相关系数为0.9799。t_(1/2)与剂量无相关性。结论在10~80 mg/kg剂量范围内木犀草素呈现线性动力学特征,剂量增加至120 mg/kg后不明显。
AIM To compare the in vivo pharmacokinetics of different doses of luteolin in rats.METHODS Rats were randomly assigned into six groups and given intragastric administration of 10,20,40,80,120 mg/kg luteolin and tail intravenous injection administration of Luteolin injection(4 mg/kg),respectively,after which blood collection was made at different time points,HPLC was adopted in the plasma concentration determination of luteolin,and main pharmacokinetic parameters were calculated.RESULTS At the dose range of 10-80 mg/kg,the correlation coefficients between AUC_(0-∞)and dose,and C_(max)and dose were 0.9966 and 0.9694,respectively.When the dose was 120 mg/kg,C_(max)was no longer increased,and the correlation coefficient between AUC 0_(~∞)and dose was 0.9799.No correlation between t_(1/2)and dose was found.CONCLUSION Luteolin demonstrates linear dynamic characteristics at the dose range of 10-80 mg/kg,which is not obvious after the dose is increased to 120 mg/kg.
