Determination of Free Cobalt Content in Methylcobalamin by Flame Atomic Absorption Spectrometry

摘要 目的建立测定甲钴胺中游离钴的分析方法。方法以大孔吸附树脂吸附甲钴胺,0.2%硝酸洗脱游离钴后,以火焰原子吸收光谱法测定洗脱液中游离钴含量。结果方法学验证结果显示,钴在0.0~3.0μg/ml呈线性,r=0.9996;钴的定量限为0.2453μg/ml,检出限为0.0736μg/ml,6份样品重复性相对标准偏差(RSD)为4.47%;甲钴胺中游离钴的加样回收率为100.9%。结论本方法游离钴测定准确率高,前处理操作简单,能够准确测定产品中的游离钴。 Objective To establish an analytical method for the determination of free cobalt in methylcobalamin.Methods The macroporous adsorption resin is used to adsorb methylcobalamin,and then the free cobalt is eluted with 0.2%nitric acid,and then the content of free cobalt in the eluate is determined by flame atomic absorption spectrometry.Results The method verification results show that the cobalt is linear in the range of 0.0~3.0μg/ml,r=0.9996.The limit of quantification of cobalt is 0.2453μg/ml,and the limit of detection is 0.07360μg/ml.The repeatability RSD of 6 samples was 4.47%.The sample recovery rate of free cobalt in methylcobalamin was 100.9%.Conclusion The method has high accuracy in determining free cobalt,simple pretreatment operation,and can accurately determine free cobalt in products.
