HPLC-DAD法同时测定养生丸中4种活性成分的含量

Simultaneous Determination of Four Active Components in Yangsheng Pills by HPLC-DAD

目的:建立高效液相色谱法同时测定医院制剂养生丸中芍药苷、阿魏酸、羌活醇、异欧前胡素4种活性指标成分的含量。方法:色谱柱Agilent Eclipse XDB-C18(150mm×4.6mm,5μm);流动相:乙腈(A)-0.1%磷酸水溶液(B),体积流量1.0mL/min;柱温30℃;样品盘温度10℃;检测波长为230nm及315nm;进样量10μL。结果:养生丸中芍药苷、阿魏酸、羌活醇、异欧前胡素的质量浓度在0.954~95.396μg·mL^(-1)(r=0.999 7)、0.307~30.68μg·mL^(-1)(r=0.999 6)、1.256~125.6μg·mL^(-1)(r=0.999 5)、0.72~72.0μg·mL^(-1)(r=0.999 5)范围内与峰面积线性关系良好,平均回收率(n=6)分别为98.89%(RSD=0.98%)、100.03%(RSD=0.74%)、101.10%(RSD=1.73%)、103.29%(RSD=0.45%)。结论:该方法经方法学验证,具有专属性强、操作简单、准确快速的优点,可用于医院制剂养生丸的质量控制。

Objective:To establish an HPLC-DAD method for the simultaneous determination of four active components including paeoniflorin,ferulic acid,notopterygium alcohol and isoimperatorin in Yangsheng Pills prepared by our hospital.Methods:Agilent Eclipse XDB-C18(150mm×4.6mm,5μm)column was used with acetonitrile(A)-0.1%phosphoric acid(B)as the mobile phase by gradient elution,at flow rate of 1.0mL/min;column temperature was maintained at 30℃;sample tray temperature was maintained at 10℃;detection wavelength was set at 230nm and 315nm,and the injection volume was 10μL.Results:The mass concentrations of paeoniflorin,ferulic acid,notopterygol and isoimperatorin had good linear relationships with peak area in the range of 0.954~95.396μg·mL^(-1)(r=0.9997),0.307~30.68μg·mL^(-1)(r=0.9996),1.256~125.6μg·mL^(-1)(r=0.9995),0.72~72.0μg·mL^(-1)(r=0.9995),and the average recoveries(n=6)were 98.89%(RSD=0.98%),100.03%(RSD=0.74%),101.10%(RSD=1.73%),103.29%(RSD=0.45%),respectively.Conclusion:The method has been verified by the method and has the advantages of strong specificity,simple operation,accuracy and rapidity.It can be used for the quality control of the Yangsheng Pills prepared by our hospital.
